1. Algorithmic discovery, development and personalized selection of higher-order drug cocktails : A label-free live-cell imaging & secretomics approach
Sammanfattning : An upward trend in clinical pharmacology is the use of multiple drugs to combat complex and co-occurring diseases due to better efficacy, decreased toxicity and reduced risk of evolving resistance. Despite high late-stage attrition rates and the need for multi drug treatments, most drug discovery and development efforts are still mainly focused on new one-size-fits-all monotherapies. LÄS MER
2. Automating STED microscopy for functional and structural live-cell imaging
Sammanfattning : Optical microscopy imaging methods are today invaluable tools for studies in life sciences as they allow visualization of biological systems, tissues, cells, and sub-cellular compartments from millimetres down to nanometres. The invention and development of nanoscopy in the past 20 years has pushed fluorescence microscopy down to the nanoscale, reaching beyond the natural diffraction limit of light that does not allow focusing of visible light below sizes of around 200 nm, and into the realm of what was previously only thought possible with electron microscopy. 3. Imaging Living Cells with an X-ray Laser
Sammanfattning : Imaging living cells at a resolution higher than the resolution of optical microscopy is a significant challenge. Fluorescence microscopy can achieve a degree of super-resolution via labeling cellular components with a fluorescent dye. LÄS MER
4. Sugar-modulated gene expression and cell division in cell culture and seedlings of A. thaliana
Sammanfattning : Throughout their life cycle, plants adjust growth in response to their developmental and environmental situation within the limits of their energetic capacities. This capacity is defined by the local sugar availability, which is constantly modulated through synthesis, transport and consumption of sugar. LÄS MER
5. Development of quantitative optical microscopy methods for single cell analysis
Sammanfattning : Optical microscopy has brought powerful methods to biological research, in particular the possibility to assay dynamic molecular processes within living individual cells. This thesis deals with various aspects of fluorescence and Raman microscopy, such as the possibility of avoiding cellular damage or stress caused by illumination, how to manipulate single cells for spectroscopic analysis and how to quantify images automatically.
