    Författare :Michael Svärd; Åke Rasmuson; Hugo Meekes; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Polymorphism; crystallization; thermodynamics; kinetics; nucleation; crystallography; solubility; phase equilibria; polymorphic transformation; solution history; metastable zone; classical theory of nucleation; two-step theory of nucleation; cluster; crystal structure prediction; lattice energy; molecular mechanics; force field; electrostatic potential; potential energy hypersurface; m-aminobenzoic acid; m-hydroxybenzoic acid; Chemical engineering; Kemiteknik;

    Sammanfattning : This work concerns the interrelationship between thermodynamic, kinetic and structural aspects of crystal polymorphism. It is both experimental and theoretical, and limited with respect to compounds to substituted monocyclic aromatics. LÄS MER

  2. 2. Structural / functional studies on human alkaline phosphatases

    Författare :Alexey Kozlenkov; Jose-Luis Millan; Hans Jörnvall; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biomedicine; mutagenesis; enzyme kinetics; computer modeling; crystallography; natural substrates; uncompetitive inhibition; isozymes; gene families; Biomedicin; Microbiology; Mikrobiologi; medicinsk genetik; Medical Genetics;

    Sammanfattning : The recent elucidation of the three-dimensional structure of human placental alkaline phosphatase (PLAP) has enabled me to perform structural studies aimed at characterizing the properties of human PLAP and tissue-nonspecific AP (TNAP) as paradigms for mammalian APs in general, using site-directed mutagenesis, protein expression, kinetic analysis and computer modeling. In Paper I, we found that a single critical E429G substitution explains the difference in stability and kinetics between the common allelic variants of PLAP and the D allozyme. LÄS MER

  3. 3. Energy Transduction and Sensory Signalling in Haloarchaea Structural Characterisation of the Photocycles of Bacteriorhodopsin and Sensory Rhodopsin II

    Författare :Karl Edman; Chalmers tekniska högskola; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; kinetic crystallography; proton pumping; Haloarchaea; structural mechanism; bacteriorhodopsin; sensory signalling; sensory rhodopsinII; retinal proteins;

    Sammanfattning : Archaeal rhodopsins belong a subfamily of heptahelical transmembrane proteins. All contain a buried retinal chromophore covalently bound to a conserved lysine residue in helix G via a protonated Schiff base. LÄS MER

  5. 5. Structural Studies of Polyamine Biosynthesis Enzymes: Potential Targets for Drugs against Tropical Parasites

    Författare :Veronica Tamu Dufe; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; inhibitors; protein crystallography; Molecular biophysics; Molekylär biofysik; tropical parasites; polyamine biosynthesis enzymes;

    Sammanfattning : Two enzymes involved in the biosynthesis of polyamines, ornithine decarboxylase (ODC, from human and Leishmania donovani) and spermidine synthase (SPDS, from Plasmodium falciparum and Caenorhabditis elegans) were studied using biochemical methods, homology modelling and X-ray crystallography. ODC catalyses the first and committed step in polyamine biosynthesis, the decarboxylation of ornithine to putrescine. LÄS MER
