  1. 1. A study of potassium channel activation as a pharmacological principle for vasodilation of cerebral blood vessels

    Författare :Torsten Ryman; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacological sciences; cerebral arteries; Cerebral ischemia; Potassium channel openers; vascular endothelium; EDHF; KRN2391; KATP; Pinacidil; pharmacognosy; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi;

    Sammanfattning : Increasing [K+]o is intimately coupled to progressive ischemia and reduced CBF. In isolated cerebral and mesenteric arteries, it was found that significant differences in the vascular responses to [K+]o exist. Among the species studied, human cerebral arteries were the most sensitive artery to increasing [K+]o. LÄS MER

  2. 2. Regulation of glucagon secretion from pancreatic alpha cells

    Författare :Yang De Marinis; diabetes och endokrinologi Genomik; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Epac2; glucagon secretion; pancreatic alpha cells; K- ATP channel; N-type Ca2 channel; L-type Ca2 channel; GLP-1; cAMP; PKA; Adrenaline; PKC;

    Sammanfattning : Glucagon secreted by pancreatic α-cells plays an important role in the regulation of blood glucose. In this thesis, different techniques such as electrophysiology, immunohistochemistry and hormone secretion assay were combined to explore the mechanisms by which glucagon secretion is regulated. 3. 3. Electrophysiology of pancreatic A-, B- and D-cells studied in intact mouse islets of Langerhans

    Författare :Sven Göpel; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; somatostatin; glucagon; insulin; patch-clamp; membrane potential; Pancreatic islet; ion channel; diabetes; Physiology; Fysiologi;

    Sammanfattning : Abstract The electrophysiological properties of insulin-secreting b-cells, glucagon-releasing a-cells and somatostation-producing d-cells in intact mouse pancreatic islets were investigated using the perforated patch whole-cell technique. It is demonstrated that it is possible to functionally identify the different cell types in situ. LÄS MER

  4. 4. Regulation of exocytosis by Ca2+ and cAMP - A study on pancreatic beta- and alpha-cells

    Författare :Xiaosong Ma; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Endocrinology; secreting systems; type-2 diabetes; SUR1; Ca2 -channel; glucagon; exocytosis; insulin; diabetology; Endokrinologi; sekretion; diabetologi;

    Sammanfattning : Type-2 diabetes is characterized by impaired insulin secretion associated with excess glucagon release. Exocytosis of insulin- or glucagon-containing granules is initiated by Ca2+-influx through voltage-dependent Ca2+-channels and is modulated by the second messengers such as cAMP. LÄS MER

  5. 5. Pulsatile insulin release from single islets of Langerhans

    Författare :Johanna Westerlund; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell biology; islets of Langerhans; insulin; ELISA; cytoplasmic calcium; oscillations; type 2 diabetes; glucose; diazoxide; potassium; tolbutamide; DNP; antimycin A; iodoacetamide; microfluorometry; fura-2; KATP channels; Cellbiologi; Cell biology; Cellbiologi; medicinsk cellbiologi; Medical Cell Biology;

    Sammanfattning : Insulin release from single islets of Langerhans is pulsatile. The secretory activities of the islets in the pancreas are coordinated resulting in plasma insulin oscillations. Nutrients amplitude-regulate the insulin pulses without influencing their frequency. LÄS MER
