1. Recreating the microenvironment of the neurovascular unit
Sammanfattning : The neurovascular unit (NVU) comprises the blood-brain-barrier (BBB) and its surrounding astrocytes, pericytes and neurons that are embedded in the extracellular matrix (ECM). As the main function of the BBB is to protect the brain from inlet of pathogens and toxins, the specialized endothelial cells that keep the barrier tight will also hinder the passage of pharmaceuticals. 2. Direct Conversion of Human Fibroblasts to Induced Neurons
Sammanfattning : During direct cellular reprogramming, forced expression of key transcription factors (TFs) directly converts one terminally differentiated cell type into that of another fate, exemplified in this theses by the conversion of fibroblasts into functional induced neurons (iNs). Direct conversion of mouse fibroblasts to functional neurons was established in 2010, and the starting point of my doctoral thesis was the aim to transfer iN-technology to human cells and to explore the potential of this technique in regards to generate subtype-specific neurons as well as to use these cells for transplantation studies. 3. Neurogenesis from Neural Stem Cells, Ependymal Cells and Fibroblasts
Sammanfattning : Stroke is a major cause of death and disability around the world. Stroke leads to loss of neurons and also other cells in the brain due to lack of blood supply. Currently no therapies are available to treat stroke-related disability. It has been shown that stroke leads to increased neurogenesis, birth of new neurons, within the brain. LÄS MER
4. Oligodendroglial (dys)function in alpha-synucleinopathies
Sammanfattning : The complexity of a-synucleinopathies, which include multiple system atrophy (MSA) and Parkinson’s disease (PD), is not entirely understood. It is known that pathological accumulation of a-synuclein (a-syn) into proteinaceous aggregates is a cellular hallmark of these diseases. LÄS MER
