  1. 1. Structure determination of Methanocaldococcus jannaschii nucleoside kinase

    Författare :Linda Arnfors; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Methanocaldococcus jannaschii; Archaea; hyperthermophile; nucleoside kinase; ribokinase family; X-ray crystallography; differential scanning calorimetry; thermal stability;

    Sammanfattning : All organisms can be divided into the tree kingdoms of life: Archaea, Bacteria and Eukarya. Archaeal organisms are often found to exhibit, compared to human perspectives, extreme environments, such as high temperature, high salt and acidic habitats. Many archaeal species are hyperthermophiles, i.e. LÄS MER

  2. 2. Molecular Simulation of Enzyme Catalysis and Inhibition

    Författare :Sinisa Bjelic; Johan Åqvist; Adrian J. Mulholland; Uppsala universitet; []
    Nyckelord :Theoretical chemistry; enzyme catalysis; enzyme inhibition; computer simulations; molecular dynamics; empirical valence bond method; structure-based inhibitor design; Teoretisk kemi;

    Sammanfattning : The reaction mechanisms for the hemoglobin degrading enzymes in the Plasmodium falciparum malaria parasite, plasmepsin II (Plm II) and histo-aspartic protease (HAP), have been analyzed by molecular simulations. The reaction free energy profiles, calculated by the empirical valence bond (EVB) method in combination with molecular dynamics (MD) and free energy perturbation (FEP) simulations are in good agreement with experimental data. 3. 3. Structural Studies of Proteins involved in Nucleotide Metabolism : Studies of a ribonucleotide reductase from A. aeolicus and NUDT15

    Författare :Daniel Rehling; Pål Stenmark; Gisela Brändén; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; X-ray crystallography; ribonucleotide reductase; NUDT15; Biochemistry; biokemi;

    Sammanfattning : This thesis is separated into two parts. The first part concerns ribonucleotide reductase from Aquifex aeolicus. A. aeolicus is a hyperthermophilic bacterium that thrives at extremely high temperatures of 80-95 °C. LÄS MER

  4. 4. Structural studies of lumazine synthases : thermostability, catalytic mechanism and molecular assembly

    Författare :Xiaofeng Zhang; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :X-ray crystallography; lumazine synthase; thermostability; catalytic mechanism; molecular assembly;

    Sammanfattning : Riboflavin, also known as vitamin B2, is biosynthesized in plants, bacteria, archaea and fungi. The primary biological function of riboflavin is related to its existence as a component of the two coenzymes, flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD), which play an important role for electron transfer in energy metabolism.
