11. Charcterization of biotransformation systems in human cells
Sammanfattning : Efforts are continuously being made to improve test procedures so that predictions of toxicity can be made earlier and more accurately. Understanding mechanisms of toxicity is an important part of improving safety assessment, but for this, simplified methods and models to screen for mechanisms associated with toxicity are required. 12. Translational genetics identifies a novel target to treat fatty liver disease
Sammanfattning : Fatty liver disease (FLD) is rapidly prevailing as the most common liver disease worldwide, with an estimated of one-quarter of the global population affected. Environmental and genetic determinants contribute to FLD susceptibility. LÄS MER
13. The role of fibroblast growth factor 19 in human liver
Sammanfattning : Bile acids are synthesized in the liver and are essential for intestinal absorption of lipids. Bile acid homeostasis is crucial and if not maintained, bile acids can accumulate to toxic levels and cause severe damage to the liver. LÄS MER
14. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury
Sammanfattning : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 15. Cyclic guanosine 3', 5'-monophosphate : studies on synthesis and efflux from hepatocytes, liver slices and cerebellar slices and on serotonin-mediated cGMP synthesis in human platelets
Sammanfattning : The present study established the existence of a carrier mediated transport of 3'5' cGMP from rat liver slices, hepatocytes and cerebellar slices in which the cellular levels of 3'5' cGMP were elevated by application of appropriate stimuli and inhibitors of the 3'5' cyclic nucleotide phosphodiesterase. The carrier is specific for 3'5' cGMP as opposed to 3'5' cAMP. LÄS MER
