  1. 1. Approaches to Optimizing High Content Confocal Microscopy

    Författare :Erland Lewin; Hjalmar Brismar; Kjell S. Carlsson; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : This thesis presents two methods for improving high contentconfocal microscopy.The author's part in the first, "Toward a confocal subcellular atlasof the human proteome" was automating image capture of foursimultaneously tagged structures in cells in 96 well plates. In total,thousands of images of hundreds of proteins in cells. LÄS MER

  2. 2. Molecular Approaches to Explore Drug-Target Interactions

    Författare :Abdullah Al-Amin; Ulf Landegren; Christopher Schofield; Uppsala universitet; []
    Nyckelord :Drug discovery; target engagement; target engagement-mediated amplification; cellular thermal shift assay; proximity extension assay; in situ PLA; high-content imaging; Molekylär medicin; Molecular Medicine;

    Sammanfattning : Improved means to assess the clinical potential of drug candidates can critically influence development of new therapeutic entities, a central aim in medical life science. Drug discovery and development relies on construction and selection of small organic compounds or biological agents that bind targets of interest. LÄS MER

  3. 3. Towards High-Throughput Phenotypic and Systemic Profiling of in vitro Growing Cell Populations using Label-Free Microscopy and Spectroscopy : Applications in Cancer Pharmacology

    Författare :Obaid Aftab; Mats Gustafsson; Neil Carragher; Uppsala universitet; []
    Nyckelord :label free vesicle detector; high-throughput; phase contrast microscopy; Library of Pharmacologically Active Compounds; High Content Screening; fluorometric microculture cytotoxicity assay; nuclear magnetic resonance; mass spectrometry;

    Sammanfattning : Modern techniques like automated microscopy and spectroscopy now make it possible to study quantitatively, across multiple phenotypic and molecular parameters, how cell populations are affected by different treatments and/or environmental disturbances. As the technology development at the instrument level often is ahead of the data analytical tools and the scientific questions, there is a large and growing need for computational algorithms enabling desired data analysis. 4. 4. Studying the cell cycle using systems biology and high content characterization of the ubiquitin proteasome system

    Författare :Johan Boström; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The cell cycle is the process through which our cells grow and divide in a carefully orchestrated and controlled manner. An uncontrolled cell cycle is a basic hallmark of cancer, in which excessive cell growth leads to adverse effects on the tissue- and organ-level. LÄS MER

  5. 5. TGFβ and LXR signaling in hepatocellular carcinoma

    Författare :Claudia Bellomo; Aristidis Moustakas; Lars Holmgren; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Hepatocellular Carcinoma; high-throughput imaging; small molecules; epithelial plasticity; LXR; Snail; TGFβ; tumor suppression; reactive oxygen species; cancer-associated fibroblast; myofibroblast; Smad3; αSMA;

    Sammanfattning : Hepatocellular carcinoma (HCC) is one of the most prevalent cancer types in the Western world and in the Asia-pacific regions, with its incidence expected to rise up to 22 million cases till 2020. Hepatocellular carcinoma etiology is mainly due to hepatitis B (HBV) and hepatitis C (HCV) infections, and to a lesser extent it is determined by the development of alcohol-driven cirrhosis and non-alcoholic steatohepatitis (NASH).
