1. Proteomics informed investigation of human hepatocytes and liver tissue
Sammanfattning : A successful drug needs to display beneficial absorption, distribution, metabolism, excretion and toxicity (ADME-Tox) profile. It is therefore important to investigate these properties during the drug discovery process. LÄS MER
2. Methylome and Transcriptome Profiling of Hepatocytes Derived from Human Pluripotent Stem Cells
Sammanfattning : Six hundred million people suffering from liver diseases worldwide of which the lethality is two million. Freshly isolated hepatocytes from the liver have been used for transplantation purposes and are extensively used to recapitulate drug metabolism. LÄS MER
3. Device Bonding and Antibody Binding: Developments for On-Chip Differentiation of Induced Pluripotent Stem Cells into Hepatocytes
Sammanfattning : Abstract This thesis describes the bonding optimization of a liver-on-a-chip microfluidic device for evaluation of cultured induced pluripotent stem cell (iPSC)-derived definitive endoderm (DE) cells during their targeted differentiation into mature hepatocytes. Furthermore, it describes the optimization of a number of antibody assays for use during said differentiation, for the purpose of studying the cellular phenotype. LÄS MER
4. Development and Applications of Microfluidic Devices for Liver-on-a-Chip Studies
Sammanfattning : Modeling the human physiology in vitro is a challenging task, yet one of importance for the development of drugs and the study of diseases. To better address the complexities of human organs, microfluidic systems called organs-on-chips are being developed to emulate organ-specific environments for cell culture. LÄS MER
5. PPARs in the regulation of gene expression and lipid metabolism in the liver
Sammanfattning : Peroxisome proliferator-activated receptors (PPARs) are ligand-activated transcription factors, regulating genes involved in carbohydrate, lipid and lipoprotein metabolism. Synthetic PPARalpha agonists used in clinic are the fibrates, and their hypotriglyceridemic effect is believed to be the result of decreased VLDL triglyceride secretion and increased turnover of triglyceride-rich lipoprotein particles in the circulation.
