  1. 1. Tryptophan-related Neurotransmission in the Brain: Disturbances Associated with Experimental Hepatic Encephalopathy

    Författare :Peter B F Bergqvist; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; tetrodotoxin; serotonin; release; rat; quinolinic acid; potassium; chloroamphetamine; noradrenaline; neurotransmission; microdialysis; hepatic encephalopathy; fenfluramine; drug safety; dopamine; coma; citalopram; ammonia; calcium; brain; tryptophan; Pharmacological sciences; pharmacognosy; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi;

    Sammanfattning : In the present study, L-tryptophan (TRP)-related disturbances in the brain in experimental hepatic encephalopathy (HE) were investigated. The endogenous NMDA-receptor agonist and L-TRP metabolite quinolinic acid (QUIN) has been suggested to be involved in the pathogenesis of HE. LÄS MER

  2. 2. Bleeding Oesophageal Varices. A Clinical Study of Mesocaval Interposition Shunting and Endoscopic Sclerotherapy

    Författare :Björn Isaksson; Malmö Institutionen för kliniska vetenskaper; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; repeated sclerotherapy; mesocaval interposition shunt; lysosomal enzymes; hepatic encephalopathy; bleeding oesophageal varices; Liver cirrhosis; portal hypertension; oesophageal manometry; Surgery; orthopaedics; traumatology; Kirurgi; ortopedi; traumatologi;

    Sammanfattning : This thesis is aimed at evaluating the clinical results of two treatments in patients with bleeding oesophageal varices; the mesocaval interposition shunt (MI shunt) and repeated endoscopic sclerotherapy (ST). 24 patients were randomised to MI shunt and 21 patients to the ST group (mean age 51.6?9.2 and 53. 3. 3. Antidepressant drug effects in vivo: Focus on pharmacokinetic and pharmacodynamic responses in different experimental paradigms

    Författare :Cecilia Wikell; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; portacaval shunt; pharmacokinetics; pharmacodynamics; hepatic encephalopathy; enantiomers; drug safety; drug disposition; citalopram; antidepressants; brain microdialysis; behaviour; venlafaxine; Pharmacological sciences; pharmacognosy; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi;

    Sammanfattning : Pharmacokinetic and pharmacodynamic activities of the antidepressants venlafaxine (VEN) and citalopram (CIT) were investigated in the portacaval shunted (PCS) rat, a model of chronic hepatic encephalopathy (HE), and normal/control rats. The levels of VEN in serum and brain were higher in PCS rats than in controls after a single injection and chronic treatment with VEN (10 mg/kg). LÄS MER

  4. 4. Experimental studies on the effects of hepatocyte-based liver support

    Författare :Helene Lilja; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Surgery; Kirurgi; Surgery; Kirurgi; Surgery; kirurgi;

    Sammanfattning : Liver transplantation is limited by lack of donors and consequently there is a need to develop liver support/replacement therapies, such as hepatocyte transplantation (HcTx) and artificial livers. In the present thesis, the effects of these therapies were investigated. First, we established a technique of isolating fetal hepatocytes (FH). LÄS MER

  5. 5. Interaction between biomaterials and innate immunity with clinical implications

    Författare :Shan Huang; Kristina Nilsson Ekdahl; Charlotte Dahle; Linnéuniversitetet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Complement system; contact system; blood; biomaterials; biocompatibility; in vitro screening; iC3; dialysis;

    Sammanfattning : Today there is an increasing clinical demand and expectation of patients for biomaterials, which underscores the importance of discovering the correlations between biomaterials and biological systems, especially blood. When an artificial material makes contact with blood, the first event is a rapid adsorption of plasma protein on the material surface, on top of which the innate immune system is triggered, with potentially detrimental consequences.
