  1. 1. Towards the Development of Photoswitchable β-Hairpin Mimetics

    Författare :Máté Erdélyi; Adolf Gogoll; Anders Karlén; John A. Robinson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; β-hairpin; peptidomimetics; Sonogashira; NMR; photochemistry; microwave; peptide; palladium; Organisk kemi; Organic chemistry; Organisk kemi;

    Sammanfattning : Peptide secondary structure mimetics are important tools in medicinal chemistry, as they provide analogues of endogeneous peptides with new physicochemical and pharmacological properties. The β-hairpin motif has been shown to be involved in numerous physiological processes, among others in regulation of eucariotic gene transcription. LÄS MER

  2. 2. Nucleic Acids as Drug Targets -Interactions of Platinum Complexes, Polyamines, and siRNA with DNA and RNA Models

    Författare :Margareta Hägerlöf; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Kinetics; Kemi; Chemistry; AU-rich region; siRNA; Circular Dichroism; Polyamine; Modified Nucleoside; HPLC; Thermodynamic Stability; cisplatin; Hairpin; DNA and RNA conformation; RNA; DNA; Anticancer Drug; Platinum Complexes;

    Sammanfattning : Drugs targeting nucleic acids were studied in DNA and RNA model systems. The effects were investigated with respect to structural and thermodynamic influence, kinetics, and biological function. The drugs employed in this thesis were anticancer active platinum(II) complexes, polyamine derivatives, and siRNA. LÄS MER

  3. 3. Activation of Herpes simplex virus type 1 origin of DNA replication

    Författare :Alireza Aslani; Göteborgs universitet; []
    Nyckelord :DNA replication; Herpes simplex virus; helicase; UL9; OBP; ICP8; ATP; oriS; hairpin;

    Sammanfattning : Herpes simplex virus type 1 has been used to study the molecular mechanism of DNA replication. Initiation of DNA replication takes place at specific regions called origins of replication. The HSV-1 genome contains three origins of replication, one copy of oriL and two copies of oriS. LÄS MER

  4. 4. Interactions of Anticancer Active Platinum(II) Complexes with DNA

    Författare :Johan Kjellström; Bröstcancer-genetik; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Inorganic chemistry; polyelectrolyte; medium effects; salt dependence; DSC; CD; HPLC; oligonucleotide; hairpin; DNA; anticancer active platinum II compounds; kinetics; mechanism; Oorganisk kemi; Cytology; oncology; cancerology; Cytologi; onkologi; cancer;

    Sammanfattning : The kinetics and mechanism for interactions of the anticancer active platinum(II) complexes cis-[PtCl2(NH3)2)], cis-[PtCl2(NH3)(c-C6H11NH2)], and trans-[PtCl2-(NH3)(quinoline)] with DNA have been studied using a combination of HPLC, circular dichroism (CD), UV/vis spectroscopy, and differential scanning calorimetry (DSC). Reactions of the monoaqua forms (charge = +1) of these complexes with single-stranded DNA oligonucleotides, typically between 13-17 bases long, were found to depend on i) the composition of the reaction medium, ii) the exact nature and location of the platination target, and iii) on the DNA structure. 5. 5. Studies on the Conformation of Transmembrane Polypeptides in Membrane Proteins

    Författare :Marika Cassel; Gunnar von Heijne; Martin Spiess; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; membrane protein; topology; transmembrane helix; helical hairpin; turn propensity; Biochemistry; Biokemi;

    Sammanfattning : The major aim of the studies that this thesis is based on has been to better define the topological determinants of the formation of so-called helical hairpins during membrane protein assembly in the ER membrane.The helical hairpin is a basic folding unit in membrane proteins.
