  1. 1. Hydrophobic Interactions in Bioseparation - Applications of Modified Green Fluorescent Protein

    Författare :Kristian Becker; Tillämpad biokemi; []
    Nyckelord :Hydrophobic interaction chromatography; capillary electrochromatography; E. coli; isolation tags; haemoglobin; green fluorescent protein; retention prediction; genetic engineering; accessible surface area;

    Sammanfattning : This thesis describes interactions that are salt-promoted and mainly based on the hydrophobicity of proteins. A series of variants of the green fluorescent protein (GFP) was first constructed by site-directed mutagenesis. Several tags with various properties were then genetically fused to the N-terminus of GFP. LÄS MER

  2. 2. Metal Binding Tags - Characterisation, Use in Bioseparation and Applications of Green Fluorescent Fusion Proteins

    Författare :Florent Bernaudat; Tillämpad biokemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Proteiner; enzymologi; enzymology; E. coli; Immobilised metal ion affinity chromatography; Affinity tags; Green fluorescent protein; Surface plasmon resonance; Genetic engineering; Screening; Proteins; Immunoassay; Biotechnology; Bioteknik;

    Sammanfattning : Most of the recombinant proteins produced nowadays are fused to affinity tags, in order to facilitate their purification through affinity chromato-graphy. Out the different tags available, poly-histidine tags are among the most commonly used and can help purification through immobilised metal ion affinity chromatography (IMAC). LÄS MER

  3. 3. Amyloid Beta – Biotherapy Target and Biotechnological Tool

    Författare :Sebastian W. Meister; Stefan Ståhl; John Löfblom; Maarten Merkx; KTH; []
    Nyckelord :Alzheimer’s disease; amyloid beta; blood-brain barrier; green-fluorescent protein; intracellular assays; proteases; protease substrate specificity; protein aggregation; protein engineering.; Bioteknologi; Biotechnology;

    Sammanfattning : Alzheimer’s disease is a progressive neurodegenerative disease characterized primarily by memory impairment and cognitive decline. As of 2020, an estimated 50 million people suffer from Alzheimer’s disease or related dementia and no disease-modifying treatment options are yet approved for clinical practice. LÄS MER

    Författare :Per-Åke Löfdahl; Per-Åke Nygren; Mikael Widersten; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Affibody molecule; β-lactamase; combinatorial library; green fluorescent protein GFP ; protein engineering; protein fragment complementation assay PCA ; selection; TEVprotease; TNF-α; Bioengineering; Bioteknik; Enzyme engineering; Enzymteknik;

    Sammanfattning : Millions of years of evolution have resulted in an immense number of different proteins, which participate in virtually every process within cells and thus are of utmost importance for allknown forms of life. In addition, there are several examples of natural proteins which have found use in applications outside their natural environment, such as the use of enzymes infood industry and washing powders or the use of antibodies in diagnostic, bioseparation or therapeutic applications. 5. 5. Peptide-tagged proteins in aqueous two-phase systems

    Författare :Anna Nilsson; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Metabolism; Biokemi; metabolism; Biochemistry; green fluorescent protein; cutinase; tryptophan; Aqueous two-phase system; peptide-tagged protein;

    Sammanfattning : This thesis deals with proteins containing peptide tags for improved partitioning in aqueous two-phase systems. Qualitatively the peptide-tagged protein partitioning could be predicted from peptide data, i.e. partitioning trends found for peptides were also found for the peptide-tagged proteins. LÄS MER
