  1. 1. Hydrophobic Interactions in Bioseparation - Applications of Modified Green Fluorescent Protein

    Författare :Kristian Becker; Tillämpad biokemi; []
    Nyckelord :Hydrophobic interaction chromatography; capillary electrochromatography; E. coli; isolation tags; haemoglobin; green fluorescent protein; retention prediction; genetic engineering; accessible surface area;

    Sammanfattning : This thesis describes interactions that are salt-promoted and mainly based on the hydrophobicity of proteins. A series of variants of the green fluorescent protein (GFP) was first constructed by site-directed mutagenesis. Several tags with various properties were then genetically fused to the N-terminus of GFP. LÄS MER

  2. 2. On bacterial formats in protein library technology

    Författare :Per-Åke Löfdahl; Per-Åke Nygren; Mikael Widersten; KTH; []
    Nyckelord :ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; Affibody molecule; β-lactamase; combinatorial library; green fluorescent protein GFP ; protein engineering; protein fragment complementation assay PCA ; selection; TEVprotease; TNF-α; Bioengineering; Bioteknik; Enzyme engineering; Enzymteknik;

    Sammanfattning : Millions of years of evolution have resulted in an immense number of different proteins, which participate in virtually every process within cells and thus are of utmost importance for allknown forms of life. In addition, there are several examples of natural proteins which have found use in applications outside their natural environment, such as the use of enzymes infood industry and washing powders or the use of antibodies in diagnostic, bioseparation or therapeutic applications. 3. 3. Amyloid Beta – Biotherapy Target and Biotechnological Tool

    Författare :Sebastian W. Meister; Stefan Ståhl; John Löfblom; Maarten Merkx; KTH; []
    Nyckelord :Alzheimer’s disease; amyloid beta; blood-brain barrier; green-fluorescent protein; intracellular assays; proteases; protease substrate specificity; protein aggregation; protein engineering.; Biotechnology; Bioteknologi;

    Sammanfattning : Alzheimer’s disease is a progressive neurodegenerative disease characterized primarily by memory impairment and cognitive decline. As of 2020, an estimated 50 million people suffer from Alzheimer’s disease or related dementia and no disease-modifying treatment options are yet approved for clinical practice. LÄS MER

  4. 4. Metal Binding Tags - Characterisation, Use in Bioseparation and Applications of Green Fluorescent Fusion Proteins

    Författare :Florent Bernaudat; Tillämpad biokemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Proteiner; enzymologi; enzymology; E. coli; Immobilised metal ion affinity chromatography; Affinity tags; Green fluorescent protein; Surface plasmon resonance; Genetic engineering; Screening; Proteins; Immunoassay; Biotechnology; Bioteknik;

    Sammanfattning : Most of the recombinant proteins produced nowadays are fused to affinity tags, in order to facilitate their purification through affinity chromato-graphy. Out the different tags available, poly-histidine tags are among the most commonly used and can help purification through immobilised metal ion affinity chromatography (IMAC). LÄS MER

  5. 5. Metal Affinity Peptides in Biotechnological Applications As tools in protein purification, protein stabilisation, metal quantification and metal tolerance of bacteria and plants

    Författare :Malin Mejàre; Tillämpad biokemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; cadmium; metal quantification; fusion tag; metal affinity tag; phage display; peptide library; metal tolerance; metal resistance; metal accumulation; bioremediation; transgenic plant; tobacco; potato; protein purification; immobilised metal affinity chromatography; metal affinity precipitation; metal detection; protein stabilisation; Bioteknik; Biotechnology; Biokemi; Metabolism; Biochemistry; metal ion;

    Sammanfattning : Metal affinity tags have been fused to proteins and demonstrated to function as tools in protein purification, protein stabilisation, quantitative metal analysis and metal tolerance of bacteria and plants. In biotechnological applications proteins frequently have to function in nonnatural environments with harsh conditions which impose requirements for high stability on the proteins. LÄS MER
