2. Transport and release of insulin granules during biphasic insulin secretion
Sammanfattning : Glucose-evoked insulin secretion exhibits two phases: The first phase represents exocytosis of insulin granules situated at the plasma membrane, whereas the second phase reflects insulin release from granules continuously recruited to the plasma membrane. This thesis investigates different aspects of phasic insulin secretion, with the specific aims to elucidate the possible contribution of insulin granule transport, as well as to study the influence of the nucleotides cAMP and NADPH to this process. 3. Nuclear Organization of Gene Expression in Adenovirus Infected Cells
Sammanfattning : Adenovirus infected cells provide a good model system for studying nuclear organization during RNA production and transport. This thesis is focused on the dynamic organization of splicing factors during the late phase of Adenovirus infection in HeLa cells, the nuclear localization of viral RNA, and the pathway used for viral RNA transport to the cytoplasm. 4. Ion transport in exocrine glands with reference to cystic fibrosis
Sammanfattning : Aspects of ion transport in rat submandibular gland acinar cells at the subcellular level, and the regulation of chloride secretion and calcium mobilization in cultured pig tracheal gland acinar cells were studied with reference to the disease cystic fibrosis.In adult rat submandibular gland acinar cells, both cholinergic and α-adrenergic stimulation induced efflux of K+ and of C1-, but the response was generally less with α-adrenergic stimulation. 5. β-cell dysfunction in Huntington's disease
Sammanfattning : Huntington's disease (HD), a fatal neurodegenerative disorder, is associated with an increased risk of diabetes mellitus. The reason for this is unknown, but considering the functional similarities of neurons and the insulin secreting pancreatic β-cell, pathological mechanisms may be shared by both cell types and account for neuronal as well as endocrine dysfunction.
