16. Glucuronyl C5-epimerases in the biosynthesis of glycosaminoglycans
Sammanfattning : This thesis is focused on two enzymes, both glucuronyl CS-epimerases, that catalyse the conversion of D-glucuronic acid (GlcA) into L-iduronic acid (IdoA) residues in the repeating disaccharide units of heparin/heparan sulphate (HS) and dermatan sulphate (DS) polymers. These glycosaminoglycans are produced as proteoglycans, in which linear polysaccharide chains are attached covalently to a protein core. LÄS MER
17. Structure and functions of heparan sulfate/heparin – Importance of glucuronyl C5-epimerase and heparanase
Sammanfattning : Heparan sulfate (HS) and heparin are linear polysaccharide chains covalently O-linked to serine residues within the core proteins, so called HS proteoglycans (PGs) or heparin PG. HSPGs are produced by almost all mammalian cells and known to play important roles in developmental processes, physiological and pathological conditions; whereas heparin PG is produced by mast cells and best known as an anticoagulant in clinic. 18. Decoding Heparan Sulfate
Sammanfattning : Heparan sulfate (HS) is a polysaccharide of glycosaminoglycan type composed of alternating hexuronic acid [either glucuronic acid (GlcA) or iduronic acid (IdoA)] and glucosamine (GlcN) units that can be sulfated in various positions. HS binds to a large number of proteins and these interactions promote many biological processes, including cell adhesion and growth factor signaling. LÄS MER
19. Regulation of Heparan Sulfate 6-O-Sulfation Patterns
Sammanfattning : Heparan sulfates (HSs) are linear, negatively charged polysaccharides composed of alternating hexuronic acid (glucuronic acid or iduronic acid) and glucosamine residues that can be substituted to varying degrees with sulfate groups. HS, localized in the extracellular matrix and on the surface of most cells, interacts with a large number of proteins. LÄS MER
20. Enzymes involved in heparan sulfate chain elongation : Function of a novel family of tumor suppressors?
Sammanfattning : Heparan sulfate (HS) is a glycosaminoglycan associated with many physiological functions such as regulation of growth factor action and cell adhesion. The HS polysaccharide is synthesized by alternating addition of D-glucuronic acid (GlcA) and N-acetyl-D-glucosamine (GlcNAc) units from the corresponding UDP-sugar precursors. LÄS MER
