1. Glucose and lipid metabolism in insulin resistance : an experimental study in fat cells
Sammanfattning : Type 2 diabetes is usually caused by a combination of pancreatic β-cell failure and insulin resistance in target tissues like liver, muscle and fat. Insulin resistance is characterised by an impaired effect of insulin to reduce hepatic glucose production and to promote glucose uptake in peripheral tissues. LÄS MER
2. Pharmacometrics Modelling in Type 2 Diabetes Mellitus : Implications on Study Design and Diabetes Disease Progression
Sammanfattning : Pharmacometric modelling is widely used in many aspects related to type 2 diabetes mellitus (T2DM), for instance in the anti-diabetes drug development, and in quantifying the disease progression of T2DM.The aim of this thesis were to improve the design of early phase anti-diabetes drug development studies with the focus on the power to identify mechanism of drug action (MoA), and to characterize and quantify the progression from prediabetes to overt diabetes, both the natural progression and the progression with diet and exercise interventions, using pharmacometrics modelling. 3. Development of Flourescence-based Immunosensors for Continous Carbohydrate Monotoring : Applications for Maltose and Glucose
Sammanfattning : Weak affinity interaction of monoclonal antibodies and carbohydrate antigens can be detected and quantified by alterations in the antibodies' intrinsic tryptophan fluorescence. These weak/transient binding events have been monitored by total internal reflection fluorescence (TlRF) by facilitating the change in intrinsic tryptophan fluorescence. LÄS MER
4. Beta-glucose 1-phosphate interconverting enzymes in Lactococcus lactis: physiological role and regulation in maltose, trehalose and glucose metabolism
Sammanfattning : Lactic acid bacteria (LAB), including Lactococcus lactis, are abundant in nature. These bacteria convert carbohydrates into mainly lactate by a rather uncomplicated metabolism. An improved understanding of the physiological and genetical mechanisms involved in carbohydrate metabolism and its regulation will improve the industrial use of LAB. LÄS MER
5. Oxidative stress and inflammation as a response to glucose exposure and dialysis
Sammanfattning : Abstract: The main player of this thesis is glucose, both on a cellular level and with a clinical approach. Too much or wrongly handled glucose contributes to increased inflammation and oxidative stress, which is reinforced by the negative influences of uraemia and dialysis treatment. LÄS MER
