1. Mechanisms of phagocytosis resistance in Streptococcus pyogenes
Sammanfattning : A distinguishing feature of Streptococcus pyogenes (GAS) is their ability to resist phagocytosis in whole human blood in the absence of type-specific antibodies, a property that is dependent on the expression of the surface associated M proteins. A common characteristic of M proteins is their interaction with a variety of host proteins including the complement regulatory protein factor H (FH) and fibrinogen that plays a key role in coagulation. 2. Novel endogenous mechanisms of complement regulation - A delicate balance
Sammanfattning : In this thesis, the biochemical mechanisms for complement activation by endogenous proteins are explored. The short leucine-rich repeat proteins (SLRPs) help organise extracellular matrices. We found that several SLRPs bind C1q and of these, fibromodulin and osteoadherin trigger complement. LÄS MER
3. Surface proteins of pathogenic streptococci
Sammanfattning : Streptococcus pyogenes (group A streptococcus) and the group B streptococcus (GBS) are two important human pathogens that cause different types of diseases and express different surface structures implicated in virulence. This thesis focuses on several surface proteins expressed by these pathogens, analyzing the biological function of these proteins and their ability to elicit protective immunity. LÄS MER
4. Structural and functional studies of C4b-binding protein (C4BP)
Sammanfattning : The subject of this thesis is complement inhibitor C4b-binding protein (C4BP), on which we have performed both structural and functional studies. Complement is part of the innate immune defence and eliminates microbes, solubilises immune complexes and is involved in the clearance of apoptotic cells. LÄS MER
5. Cancer and autoimmunity: two sides of the same coin. Complement system proteins in the regulation of immune responses
Sammanfattning : Factor H is an inhibitor of the complement system, essential for controlling the alternative pathway and also playing a crucial role in various cellular functions. In this thesis, we identified new intriguing functions of FH as a survival factor for primary human monocytes, promoting their differentiation into immunosuppressive macrophages. LÄS MER
