16. How can p53 induce differentiation of leukemic cells?
Sammanfattning : The terminal differentiation of leukemic cells is closely connected to their death. The concept of differentiation therapy originates from the idea that forced maturation of leukemic cells could induce cell death without the side-effects that are provoked by ordinary chemotherapy. 17. Characterization of human myeloid progenitors and their differentiation
Sammanfattning : All hematopoietic cells are derived from the hematopoietic stem cells (HSCs) in a continuous and dynamic process, regulated by a network of transcription factors in different combinations and extrinsic signals from growth factors (GFs) and other factors in the microenvironment. In the initial stages of hematopoiesis HSCs differentiate to progenitor cells with gradually more restricted lineage potential and according to the most accepted model the first restriction in developmental potential results in commitment to either the lymphoid or the myeloid branch, in the latter followed by commitment to the neutrophil/monocyte and erythrocyte/megakaryocyte lineages. 18. On the Role of the Tumor Suppressor p53 in Leukemic Cell Differentiation
Sammanfattning : Leukemic cells suffer from an impaired ability to differentiate due to inherited or acquired genetic lesions. These genetic changes can sometimes be bypassed with various compounds both in vitro, and, more rarely, in vivo, thus inducing terminal differentiation of the leukemic cells. LÄS MER
19. Molecular Genetic Studies of Ph-positive Leukemias and the BCR and ABL Genes
Sammanfattning : In the present thesis molecular genetic methods were used to address different biological and clinical aspects of Philadelphia (Ph) chromosome-positive leukemias and the BCR and ABL genes. In the first study, children with Ph-positive chronic myeloid leukemia (CML) were found to have essentially the same breakpoint locations in BCR as adults, a pattern which differs from the one seen in Ph-positive acute lymphoid leukemia (ALL). 20. Diamond-Blackfan Anemia: Erythropoiesis Lost in Translation
Sammanfattning : Diamond-Blackfan anemia (DBA) is a congenital erythroid aplasia that usually presents as macrocytic anemia during infancy. Linkage analysis suggests that at least four genes are associated with DBA of which two have been identified so far. LÄS MER
