  1. 1. Biopharmaceutical aspects of intestinal drug absorption : Regional permeability and absorption-modifying excipients

    Författare :David Dahlgren; Hans Lennernäs; Erik Sjögren; Bertil Abrahamsson; Christer Tannergren; Steffansen Bente; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intestinal permeability; absorption-modifying excipients; Biofarmaci; Biopharmaceutics;

    Sammanfattning : Before an orally administered drug reaches the systemic circulation, it has to dissolve in the intestinal fluids, permeate across the intestinal epithelial cell barrier, and pass through the liver. The permeation rate of drug compounds can be low and show regional differences.The thesis had two general aims. LÄS MER

  2. 2. Multivariate methods in tablet formulation

    Författare :Jon Gabrielsson; Michael Sjöström; Svante Wold; Torbjörn Lundstedt; Marc Brown; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; PCA; statistical experimental design; multivariate design; PLS; excipients; direct compression; tablet formulation; robustness testing; Organisk kemi; Organic chemistry; Organisk kemi; organisk kemi; Organic Chemistry;

    Sammanfattning : This thesis describes the application of multivariate methods in a novel approach to the formulation of tablets for direct compression. It begins with a brief historical review, followed by a basic introduction to key aspects of tablet formulation and multivariate data analysis. LÄS MER

  3. 3. Interfacial and material aspects of powders with relevance to pharmaceutical tableting performance

    Författare :Maria Badal Tejedor; Mark W. Rutland; William Ducker; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; atomic force microscopy; excipients; surface characterization; tableting; milling; amorphisation; Kemi; Chemistry;

    Sammanfattning : Tablets are the most common forms of drug administration. They are convenient to administer and easy to manufacture. However, problems associated with the adhesion of the powders to the tableting tools are common. This phenomenon is known as sticking and even though it has been well documented and studied, it remains poorly understood. LÄS MER

  4. 4. Intestinal absorption of drugs : The impact of regional permeability, nanoparticles, and absorption-modifying excipients

    Författare :Carl Roos; Hans Lennernäs; Erik Sjögren; Martin Brandl; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Permeability; intestinal absorption; regional permeability; nanoparticles; nanosuspensions; absorption-modifying excipients; AMEs; Biofarmaci; Biopharmaceutics;

    Sammanfattning : For successful delivery of orally given drug products, the drug compounds must have adequate solubility and permeability in the human gastrointestinal tract. The permeability of a compound is determined by its size and lipophilicity, and is usually evaluated in various pre-clinical models, including rat models. LÄS MER

  5. 5. Dissolving the Rocks : Solubility Enhancement of Active Pharmaceutical Ingredients using Mesoporous Silica

    Författare :Xin Xia; Alfonso E. Garcia-Bennett; Xiaodong Zou; Göran Frenning; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; mesoporous silica; drug delivery; solubility enhancement; active pharmaceutical ingredients; oral toxicity; confinement; crystallization; pharmaceutical excipients; bioavailability; Inorganic Chemistry; oorganisk kemi;

    Sammanfattning : Poor aqueous solubility is one of the greatest barriers for new drug candidates to enter toxicology studies, let alone clinical trials. This thesis focuses on contributing to solving this problem, evaluating the oral toxicity of mesoporous silica particles, and enhancing the apparent solubility and bioavailability of active pharmaceutical ingredients in vitro and in vivo using mesoporous silica particles.
