1. Methods for Preclinical Evaluation of Cytotoxic Drugs : With Special Reference to the Cyanoguanidine CHS 828 and Hollow Fiber Method
Sammanfattning : The novel cyanoguanidine CHS 828 has shown promising antitumor activity in many in vitro and in vivo studies. The long-term 14 days in vitro hollow fiber cultures, where tumor cells from different tumor cell lines were cultured inside semipermeable fibers, were more resistant to CHS 828 and other cytotoxic drugs than the shorter-term 3 days cultures. 2. Pluripotent Stem Cells of Embryonic Origin : Applications in Developmental Toxicology
Sammanfattning : General toxicity evaluation and risk assessment for human exposure is essential when developing new pharmaceuticals and chemicals. Developmental toxicology is an important part of this risk assessment which consumes large resources and many laboratory animals. LÄS MER
3. Developmental Neurotoxicity Testing Using In vitro Approaches
Sammanfattning : There is a great concern about children’s health as the developing brain in foetuses and children is much more vulnerable to injury caused by different classes of chemicals than the adult brain. This vulnerability is partly due to the fact that the adult brain is well protected against chemicals by the blood brain barrier (BBB) and children have increased absorption rates and diminished ability to detoxify many exogenous compounds, in comparison to that of adults. 4. Human liver in vitro models for evaluation of drug metabolism and disposition
Sammanfattning : The administrated dose of a drug is adjusted to give a therapeutic effect in patients without causing side-effects or toxicity. Cytochrome P450 (P450) and UDP-glucuronosyltransferase (UGT) enzymes, uptake and efflux transporters and nuclear receptors regulating these enzymes, expressed in the liver and in other tissues, are all important players in drug metabolism, disposition and elimination. 5. Calcium-induced mitochondrial permeability transition in CNS-derived mitochondria - Pharmacological aspects of specificity and toxicity in neuroprotection
Sammanfattning : Mitochondria are the main site for energy conversion in the cell. Under certain circumstances, such as excess calcium retention or production of reactive oxygen species, a pore forms at contact sites between the outer and the normally impermeable inner mitochondrial membrane.
