1. Autoantibodies and the Type I Interferon System in the Etiopathogenesis of Systemic Lupus Erythematosus
Sammanfattning : In sera remitted for anti-nuclear antibody (ANA) analysis, the supplement of a sensitive anti-SSA/Ro ELISA to the conventional ANA screening by immunofluorescence (IF) revealed that one fourth of the individuals with IF-ANA negative, but SSA/Ro ELISA positive sera, had systemic lupus erythematosus (SLE) or cutaneous LE. Consequently, adding a sensitive anti-SSA/Ro ELISA to the ANA screening is valuable for the serological detection of ANA negative SLE/LE patients. LÄS MER
2. RNA in model lipid membranes - interactions in bulk and at surfaces
Sammanfattning : The work presented in the thesis “RNA in model lipid membranes- interactions in bulk and at surfaces” aims to characterize the interaction between RNA and model membranes with different properties as well as to determine if ssRNA associate with model membranes in different ways. Several techniques have been used to characterize these systems, including quartz crystal microbalance with dissipation monitoring (QCM-D), Langmuir film balance in combination with neutron reflectometry and fluorescence microscopy as well as differential scanning calorimetry (DSC) and isothermal titration calorimetry (ITC). 3. Structural Studies of Large dsDNA Viruses using Single Particle Methods
Sammanfattning : Structural studies of large biological assemblies pose a unique problem due to their size, complexity and heterogeneity. Conventional methods like x-ray crystallography, NMR, etc. are limited in their ability to address these issues. To overcome some of these limitations, single particle methods were used. LÄS MER
4. Homing Endonucleases and Horizontal Gene Transfer in Bacteria and Bacteriophages
Sammanfattning : Homing endonuclease genes (HEGs) are selfish genetic elements that mediate their own super-Mendelian inheritance. This is mediated by the homing endonuclease cleavage of a HEG- allele followed by recombination-repair with a HEG+ allele. The majority of the HEGs are encoded in intervening sequences (IVSs). LÄS MER
5. Studies on anti-dsDNA Antibodies and other potential biomarkers in Systemic Lupus Erythematosus
Sammanfattning : Systemic Lupus Erythematosus (SLE) is characterized by chronic inflammation involving many organ systems. The etiology is unknown and the pathogenesis has not been fully elucidated. Impairments of the immune system and of the mechanisms regulating cell death are commonly seen in SLE patients. LÄS MER
