  1. 21. Accelerating the discovery of drugs for Neglected Tropical Diseases using biophysical methods

    Författare :Giulia Opassi; Helena Danielson; Jenny Sandmark; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Neglected Tropical Diseases; Drug Discovery; Biochemistry; Biophysics; Kinetic; Fragment Based Lead Discovery; Assay Development;

    Sammanfattning : Even though they account for 10% of the global disease burden and besides the new record of funding in 2018, tropical diseases are still neglected by the majority of pharmaceutical companies and public funding. The reason can be mostly found in the fact that for these diseases a conventional drug discovery process is often too expensive. LÄS MER

  2. 22. New Paradigms in GPCR Drug Discovery : Structure Prediction and Design of Ligands with Tailored Properties

    Författare :Mariama Jaiteh; Jens Carlsson; David E Gloriam; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; G Protein-Coupled Receptor; Molecular Docking; Virtual Screening; Homology Modeling; Molecular Dynamics Simulation; Chemical Library; Functionally Selective Ligand; Polypharmacology; Dimerization; Biology with specialization in Molecular Biotechnology; Biologi med inriktning mot molekylär bioteknik;

    Sammanfattning : G protein-coupled receptors (GPCRs) constitute a large superfamily of membrane proteins with key roles in cellular signaling. Upon activation by a ligand, GPCRs transduce signals from the extracellular to the intracellular environment. LÄS MER

  3. 23. Fragment Based Drug Discovery with Surface Plasmon Resonance Technology

    Författare :Helena Nordström; Helena Danielson; Gregg Siegal; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Fragment based drug discovery; SPR biosensor; matrix metalloproteinase-12; HIV-1 protease; resistance;

    Sammanfattning : Fragment based drug discovery (FBDD) has been applied to two protease drug targets, MMP-12 and HIV-1 protease. The primary screening and characterization of hit fragments were performed with surface plasmon resonance -technology. Further evaluation of the interaction was done by inhibition studies and in one case with X-ray crystallography. LÄS MER

  4. 24. Computational and Experimental Models for the Prediction of Intestinal Drug Solubility and Absorption

    Författare :Christel A. S. Bergström; Per Artursson; Han van de Waterbeemd; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutics; aqueous drug solubility; melting point; intestinal drug permeability; pH-dependent solubility; multivariate data analysis; molecular descriptors; molecular surface area; in silico modeling; drug absorption; Galenisk farmaci; Pharmaceutics; Galenisk farmaci;

    Sammanfattning : New effective experimental techniques in medicinal chemistry and pharmacology have resulted in a vast increase in the number of pharmacologically interesting compounds. However, the number of new drugs undergoing clinical trial has not augmented at the same pace, which in part has been attributed to poor absorption of the compounds. LÄS MER

  5. 25. Algorithmic discovery, development and personalized selection of higher-order drug cocktails : A label-free live-cell imaging & secretomics approach

    Författare :Efthymia Chantzi; Mats G Gustafsson; Ulf Hammerling; Andreas Bender; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug combination discovery and development; personalized pharmacotherapy; higher-order drug combination analysis; quantitative label-free live-cell imaging; secretomics; cell-cell communication; deep learning; convolutional neural networks; data mining; MapReduce; resampling; generalized highest single agent; COMBImageDL; COMBSecretomics; Bioinformatics; Bioinformatik; Pharmacology; Farmakologi; Engineering Science; Teknisk fysik;

    Sammanfattning : An upward trend in clinical pharmacology is the use of multiple drugs to combat complex and co-occurring diseases due to better efficacy, decreased toxicity and reduced risk of evolving resistance. Despite high late-stage attrition rates and the need for multi drug treatments, most drug discovery and development efforts are still mainly focused on new one-size-fits-all monotherapies. LÄS MER
