11. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development
Sammanfattning : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 12. 3D Printed scaffolds as cancer microenvironment medels for drug discovery
Sammanfattning : Cancer is one of the most common diseases in the modern world and major efforts are made globally to develop new diagnostics and treatments. It originates from a cell which, at some point, has begun to divide and grow uncontrollably. The most common type is breast cancer, and like all other cancers there is a need of more efficient drug therapies. LÄS MER
13. Algorithmic discovery, development and personalized selection of higher-order drug cocktails : A label-free live-cell imaging & secretomics approach
Sammanfattning : An upward trend in clinical pharmacology is the use of multiple drugs to combat complex and co-occurring diseases due to better efficacy, decreased toxicity and reduced risk of evolving resistance. Despite high late-stage attrition rates and the need for multi drug treatments, most drug discovery and development efforts are still mainly focused on new one-size-fits-all monotherapies. LÄS MER
14. Molecular simulations of G protein-coupled receptors : A journey into structure-based ligand design and receptor function
Sammanfattning : The superfamily of G protein-coupled receptors (GPCRs) contains a large number of important drug targets. These cell surface receptors recognize extracellular signaling molecules, which stimulates intracellular pathways that play major roles in human physiology. LÄS MER
15. Exploring Inhibitors of HIV-1 Protease : Interaction Studies with Applications for Drug Discovery
Sammanfattning : A variety of HIV-1 protease inhibitors and their interactions with the enzyme have been characterized in order to identify novel and improved drugs against AIDS. The investigated inhibitors were represented by clinical and non-clinical inhibitors, active site and allosteric inhibitors, transition-state analogues and metal-ions. LÄS MER
