  1. 1. Drug Analysis : Bioanalytical Method Development and Validation

    Författare :Mikaela Malm; Yngve Bergqvist; Anders Colmsjö; Uppsala universitet; []
    Nyckelord :Analytical chemistry; African trypanosomiasis; calibration model; chiral chromatography; dried blood spots; eflornithine; evaporative light-scattering detection; HIV AIDS; lamivudine; liquid chromatography; malaria; nevirapine; piperaquine; pneumonia; solid-phase extraction; sulfadoxine; sulfamethoxazole; validation; zidovudine; Analytisk kemi;

    Sammanfattning : This thesis describes bioanalytical methods for drug determination in biological matrixes, with drugs in focus used against diseases largely affecting low-income countries. Solid-phase extraction is used for sample cleanup, and processed samples are analyzed by liquid chromatography. LÄS MER

  2. 2. Circumventing drug resistance : Studies exploring the expediency of a total cell kill chemoresistance assay

    Författare :Bertil Jonsson; Uppsala universitet; []
    Nyckelord :Medical sciences; drug resistance; drug resistance assay; clinical correlations; cancer; human; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; klinisk farmakologi; Clinical Pharmacology;

    Sammanfattning : Cellular resistance to anti-cancer drugs is considered to be the major cause of treatment failure in clinical practice. The present studies have therefore focused on ways to circumvent resistance as evaluated by an in vitro chemoresistance assay. LÄS MER

  3. 3. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development

    Författare :Matthis Geitmann; U. Helena Danielson; Marc H. V. van Regenmortel; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; SPR biosensor; HIV-1 reverse transcriptase; HCMV protease; interaction kinetics; drug discovery; non-nucleoside inhibitor; resistance; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 4. 4. Development and characterization of an in vitro method for interaction studies between polymers and pharmaceuticals : Aiding in the development of new drug delivery systems

    Författare :Marcus Wanselius; Per Hansson; Magnus Bergström; Susanna Abrahmsén-Alami; Christian Pedersen; Anna Fureby; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Microgels; Subcutaneous administration; Drug delivery; Microfluidics; In vitro methodology; Method development; Biopharmaceuticals; Amphiphilic drugs; Polyelectrolytes; Hyaluronic acid; Peptides; Farmaceutisk fysikalisk kemi; Pharmaceutical Physical Chemistry;

    Sammanfattning : Polymers are a group of macromolecules used in formulations of pharmaceuticals, one example being the delivery system DC Bead™. Further, some of the most abundant and for drug delivery important constituents of the subcutaneous tissue are charged polymers (polyelectrolytes), e.g. collagen, hyaluronic acid, and chondroitin sulfate. 5. 5. Pharmacometric tools to support translational drug development

    Författare :Rami Ayoun Alsoud; Ulrika S. H. Simonsson; Michael Lyons; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug development; pharmacometrics; pharmacokinetics; pharmacodynamics; non-linear mixed effect models; tuberculosis; dose selection; interspecies scaling; pediatric trials; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : The use of model-informed drug development has been shown to save significant costs and improve decision making early in the drug development process. The work in this PhD thesis aimed to employ pharmacometric tools to support translational drug development from the preclinical to the late clinical stages. LÄS MER
