  1. 11. Direct Thrombin Inhibitors in Treatment and Prevention of Venous Thromboembolism: Dose – Concentration – Response Relationships

    Författare :Marie Cullberg; Mats O. Karlsson; Ulf G. Eriksson; Nick H.G. Holford; Uppsala universitet; []
    Nyckelord :Pharmaceutical biosciences; Ximelagatran; pharmacokinetic; pharmacodynamic; activated partial thromboplastin time; utility function; dosing strategy; venous thromboembolism; NONMEM; Farmaceutisk biovetenskap;

    Sammanfattning : For prevention and treatment of thrombotic diseases with an anticoagulant drug it is important that an adequate dose is given to avoid occurrence or recurrence of thrombosis, without increasing the risk of bleeding and other adverse events to unacceptable levels. The aim of this thesis was to develop mathematical models that describe the dose-concentration (pharmacokinetic) and concentration-response (pharmacodynamic) relationships of direct thrombin inhibitors, in order to estimate optimal dosages for treatment and long-term secondary prevention of venous thromboembolism (VTE). 2. 12. Modelling and Simulation to Improve Antimalarial Therapy

    Författare :Jesmin Permala Lohy Das; Mats Karlsson; Martin Bergstrand; Joel Tarning; Mads Krielgaard; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacometrics; pharmacokinetics; pharmacodynamics; malaria; artemisinin; weekly dosing; resistant; pregnant populations; intermittent preventive therapy; parasite clearance; day 3 positivity; nomogram; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Sammanfattning : The introduction of artemisinin-based combination therapy (ACT) substantially reduced malaria-related mortality and morbidity during the past decade. Despite the widespread use of ACT, there is still a considerable knowledge gap with regards to safety, efficacy and pharmacokinetic properties of these drugs, particularly in vulnerable populations like children and pregnant women. 3. 13. Contaminated sediments: Methods to assess release and toxicity of organic chemical mixtures

    Författare :Lukas Mustajärvi; Anna Sobek; Jarkko Akkanen; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Sediment; Hydrophobic organic contaminants; Flux; Bioturbation; Passive sampling; Passive dosing; Mixture toxicity; Applied Environmental Science; tillämpad miljövetenskap;

    Sammanfattning : Bottom sediments around the world store large amounts of legacy hydrophobic organic contaminants (HOCs), forming mixtures of unknown chemical composition. Primary emissions to the environment of many HOCs have been reduced as a consequence of regulation. LÄS MER

  4. 14. Chlorinated Paraffins: improved understanding of their bioaccumulation and toxicity in Daphnia magna

    Författare :Mafalda Castro; Magnus Breitholtz; Anna Sobek; Elena Gorokhova; Lillemor Asplund; Martin Scheringer; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; chlorinated paraffins; POPs; bioaccumulation; toxicity; ecotoxicology; aquatic toxicology; daphnia magna; liposomes; passive dosing; hydrophobic; mixtures; Applied Environmental Science; tillämpad miljövetenskap;

    Sammanfattning : Chlorinated paraffins (CPs) are industrial chemicals, mainly used as flame retardants, plasticizers and metal cutting fluids. Their production has reached historically high levels in the last decade, with an annual production exceeding one million tonnes. LÄS MER

  5. 15. Optimization of Colistin Dosage in the Treatment of Multiresistant Gram-negative Infections

    Författare :Matti Karvanen; Otto Cars; Lena E Friberg; Vincent H Tam; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Colistin; CMS; Pharmacology; Pharmacokinetics; PKPD; Antibiotics; Combination therapy; Pharmacometrics; Dosing regimens; Antibiotic resistance; Infectious Diseases; Infektionssjukdomar;

    Sammanfattning : As multidrug resistance in Gram-negative bacilli increases, the old antibiotic colistin has rapidly gained attention as one of few last line treatment options in the form of colistin methanesulfonate (CMS), which is hydrolyzed to colistin both in vitro and in vivo. There is a dearth of knowledge on fundamental aspects of colistin, including pharmacokinetics and optimal dosing regimens. LÄS MER
