  1. 11. Positron emission tomography in serotonergic drug development

    Författare :Bengt Andrée; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :PET; 5-HT1A receptors; 5-HT2A receptors; D2 dopamine receptors; antidepressive drugs; antipsychotic drugs; pindolol; NAD-299; S 16924; ORG 5222; M100907; WAY; DWAY;

    Sammanfattning : The serotonin neurotransmission system has a central role in drug treatment of depression and anxiety and is also discussed in relation to treatment of schizophrenia. The benefits of presently available drugs are however limited by partial of poor response and side effects. LÄS MER

  2. 12. Clinical molecular imaging of schizophrenia

    Författare :Mirjam Talvik; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :PET; D2 D3 receptors; 5HT2A receptors; antipsychotic drugs; schizophrenia; age effects; raclopride; FLB 457; M100907; clozapine; haloperidol; perphenazine; thalamus; cortex; striatum; PANSS;

    Sammanfattning : The aim of this thesis was to examine pathophysiology and antipsychotic treatment of schizophrenia by using positron emission tomography (PET). The high affinity radioligand [11C]FLB 457 was used for determination of dopamine D2 receptor binding in the thalamus and the cortex of nine drug-naïve patients with schizophrenia and eight control subjects. 3. 13. PET studies on the mechanism of monoaminergic drugs

    Författare :Magdalena Nord; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Positron emission tomography (PET) is a molecular imaging technique that allows for examinations of neurochemistry directly in the living human brain. In the present thesis, PET was used to assess the pharmacological effects of two drugs representing the major classes of antipsychotic and antidepressant drugs. LÄS MER

  4. 14. Utility of combined treatment with antipsychotic and antidepressant drugs : scientific rationales and clinical implications

    Författare :Carl Björkholm; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The atypical antipsychotic drug (APD) clozapine is the most efficacious APD in treatment-resistant schizophrenia including negative symptoms and cognitive impairment, and still lacks extrapyramidal side effects (EPS). Clozapine, which also possesses an antidepressant effect and can be used as monotherapy in bipolar disorder, has a broad receptor binding profile with higher affinity for the α2-adrenoceptor and several serotonergic receptors than D2 receptor, enhances dopamine output in the medial prefrontal cortex (mPFC) and facilitates glutamatergic NMDA receptor-mediated transmission in pyramidal cells in the same brain region. 5. 15. Novel pharmacological treatment alternatives for schizophrenia

    Författare :Charlotte Wiker; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Antipsychotic drugs are commonly divided into typical (first generation) and atypical (second generation) antipsychotic drugs. Clinically effective doses of typical antipsychotic drugs generate a striatal dopamine D2 receptor occupancy of about 70 80%, approaching a level that is associated with a high risk of extrapyramidal side effects.
