  1. 1. Functional brain activity in Alzheimer patients as studied by multi-tracer positron emission tomography : effects of treatment with cholinesterase inhibitors

    Författare :Ahmadul Kadir; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Alzheimer’s disease; cholinesterase inhibitors; positron emission tomography; nicotinic acetylcholine receptor; acetylcholinesterase; multi-tracer; 11C-nicotine; 11C-PMP; 18F-FDG; 11C-PIB; donepezil; rivastigmine; galantamine; phenserine.;

    Sammanfattning : Alzheimer’s disease (AD) is a progressive neurodegenerative disease accompanied by cognitive impairment and disturbances in several neurotransmitter systems, especially the cholinergic system. Studies have correlated the cognitive impairment observed in AD patients with a deficit in central cholinergic neurotransmission. LÄS MER

  2. 2. Long Term Outcome and Prediction Models of Cognition, Activities of Daily Living and Nursing Home Placement in Alzheimer’s Disease with Cholinesterase Inhibitor Treatment

    Författare :Carina Wattmo; Klinisk minnesforskning; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; longitudinal studies; nursing home placement; activities of daily living; cognition; Alzheimer s disease; cholinesterase inhibitors; disease progression; predictors; statistical models.;

    Sammanfattning : Background Prospective longitudinal studies in Alzheimer's disease (AD) that include cholinesterase inhibitor (ChEI) treatment in routine clinical settings are scarce. The patients vary in severity of the disease, clinical course, rate of progression and response to treatment. LÄS MER

  3. 3. Cholinesterase inhibitors in Alzheimer's disease : an experimental study on mechanisms of interatcion with muscarinic and nicotinic receptors and neuroprotection

    Författare :Anne-Lie Svensson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Alzheimer s disease Cholinesterase inhibitors Muscarinic receptors Nicotinic receptors Acetylcholine release Neuroprotection B-amyloid apolipoprotein E;

    Sammanfattning : The overall aim of this thesis was to investigate the mechanisms beyond the interaction of cholinesterase inhibitors (ChEIs) with the cholinergic system in brain, with emphasis on their regulatory and mechanistic effects on subtypes of muscarinic and nicotinic receptors and neuroprotective properties. To investigate muscarinic receptor subtypes in frontal cortices of AD brains, the muscarinic antagonists pirenzepine, AF-DX 116, HHSiD, p-F-HHSiD and himbacine were used in receptor binding assays. 4. 4. Modulation of acetylcholine release by serotonergic 5-HT1A and 5-HT1B receptors : a microdialysis study in the awake rat

    Författare :Xiao Jing Hu; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Microdialysis; Acetylcholine; Serotonin; 5-HT1A receptor; 5-HT1B receptor; Frontal cortex; Ventral hippocampus;

    Sammanfattning : The aim of the thesis was to investigate the effects of the 5-HT1A receptor antagonist robalzotan (NAD-299) and the 5-HT1B receptor antagonist NAS-181 ((R)-(+)-2-(3-morpholinomethyl-2Hchromen-8-yl) oxymethyl-morpholine methanesulfonate) on cholinergic, glutamatergic and GABAergic neurotransmission in the rat brain in vivo. The extracellular levels of acetylcholine (ACh), glutamate (Glu) and GABA were monitored by microdialysis in the frontal cortex (FC) and ventral hippocampus (VHipp) in separate groups of awake rats. 5. 5. Economic evaluation of treatments for Alzheimer's disease

    Författare :Linus Jönsson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Dementia; costs; quality of life; caregivers;

    Sammanfattning : Alzheimer's disease (AD) is the most common cause of dementia, leading to progressive cognitive impairment and loss of functional abilities for die afflicted patient, a heavy burden on caregivers and high costs for society. New treatment strategies are being introduced setting focus on the need to make informed decisions on the allocation of resources to and within dementia care. LÄS MER
