6. Protein Dynamics Studied by NMR. Kinetics of the Adipocyte Fatty Acid-Binding Protein and Oligomerisation of the Low Molecular Weight Protein Tyrosine Phosphatase
Sammanfattning : Nuclear magnetic resonance spectroscopy (NMR) has been used to determine the kinetics of ligand binding to the adipocyte fatty acid-binding protein (A-FABP), as well as the oligomerisation and inter- and intramolecular dynamics of the low molecular weight-protein tyrosine phosphatase (LMW-PTP) in solution. NMR spectroscopy is sensitive to dynamic processes on timescales ranging from picoseconds to days. LÄS MER
7. Macrolide Antibiotics in Bacterial Protein Synthesis
Sammanfattning : Macrolides are a large group of clinically relevant antibiotics that inhibit protein synthesis by binding to the large ribosomal subunit in the peptide exit tunnel, close to the peptidyl transferase center (PTC). We have shown that the peptide length of the resulting peptidyl-tRNA drop-off products is proportional to the distance between the PTC and the respective macrolide in the tunnel. LÄS MER
8. Interaction Characteristics of Viral Protease Targets and Inhibitors : Perspectives for drug discovery and development of model systems
Sammanfattning : Viral proteases are important targets for anti-viral drugs. Discovery of protease inhibitors as anti-viral drugs is aided by an understanding of the interactions between viral protease and inhibitors. This thesis addresses the characterization of protease-inhibitor interactions for application to drug discovery and model system development. LÄS MER
9. DNA Interaction Dynamics of Ruthenium(II) Complexes
Sammanfattning : DNA-binding of small synthetic ligands is of great importance in a number of different applications, from medicine to molecular biology. This thesis focuses on interactions between DNA and ruthenium(II) complexes, in particular the dimers [µ-bidppz(L)4Ru2]4+ (L=1,10-phenanthroline (P) or L=2,2-bipyridine (B)). LÄS MER
10. Development of Flourescence-based Immunosensors for Continous Carbohydrate Monotoring : Applications for Maltose and Glucose
Sammanfattning : Weak affinity interaction of monoclonal antibodies and carbohydrate antigens can be detected and quantified by alterations in the antibodies' intrinsic tryptophan fluorescence. These weak/transient binding events have been monitored by total internal reflection fluorescence (TlRF) by facilitating the change in intrinsic tryptophan fluorescence. LÄS MER
