  1. 1. Studies on Cytotoxic and Neutrophil Challenging Polypeptides and Cardiac Glycosides of Plant Origin

    Författare :Senia Johansson; Tore Mellstrand; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical chemistry; cytotoxic; neutrophil; polypeptides; Phoradendron tomentosum; Digitalis purpurea; plant; mistletoe; MS MS; Edman degradation; breast cancer; macrocyclic peptides; multidrug resistance protein; primary structures; phoratoxin; isolation; digitoxin; cardiac lycoside; structure-activity; Farmaceutisk kemi; Pharmaceutical chemistry; Farmaceutisk kemi; Pharmacognosy; farmakognosi;

    Sammanfattning : This thesis examines the isolation and characterisation (biological and chemical) of polypeptides from plants. A fractionation protocol was developed and applied on 100 plant materials with the aim of isolating highly purified polypeptide fractions from small amounts of plant materials. LÄS MER

  2. 2. Cell-penetrating peptides : Uptake, stability and biological activity

    Författare :Tina Holm; Ülo Langel; Roland Brock; Stockholms universitet; []
    Nyckelord :NATURAL SCIENCES; NATURVETENSKAP; Neurochemistry and Neurotoxicology; neurokemi och neurotoxikologi;

    Sammanfattning : Cell-penetrating peptides (CPPs) have emerged as a group of remarkable delivery vectors for various hydrophilic macromolecules, otherwise excluded from cells due to the protective plasma membrane. Unbiased conclusions regarding e.g. LÄS MER

  3. 3. Distribution and Chemical Diversity of Cyclotides from Violaceae : Impact of Structure on Cytotoxic Activity and Membrane Interactions

    Författare :Robert Burman; Ulf Göransson; Lars Bohlin; William Gerwick; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cyclotide; Violaceae; Viola; Cytotoxic; Mechanism; In vivo; Structure-activity; Membrane; Mass spectrometry; Pharmacognosy; Farmakognosi; Pharmacognosy; Farmakognosi;

    Sammanfattning : During the last decade there has been increased interest in the cyclotide protein family, which consist of a circular chain of approximately 30 amino acids, including six cysteines that form three disulfide bonds, arranged in a cyclic cystine knot motif. This thesis gives new insights in cyclotide distribution and occurrence in the plant family Violaceae, structure-activity relationships for cytotoxic effects, membrane disruption and adsorption on lipid membranes, and evaluates toxicity and anti-tumor activity in vivo. 4. 4. Vascular actions of antimicrobial peptides

    Författare :Cristina Ciornei; Sektion II; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Kardiovaskulära systemet; Cardiovascular system; vascular smooth muscle; sepsis; nitric oxide; human antimicrobial peptide LL-37; Antimicrobial peptides; lipopolysaccharide; bactericidal permeability-increasing protein;

    Sammanfattning : The present thesis focuses on the vascular effects of antimicrobial peptides especially mechanisms involving the vasodilator nitric oxide (NO), which is released from the blood vessel wall during inflammatory conditions such as sepsis. Bacterial lipopolysaccharide stimulates the expression of nitric oxide synthase (iNOS), which increases the NO production in vascular tissues or cultured vascular smooth muscle cells (VSMC). LÄS MER

  5. 5. Actions of Antimicrobial Peptides and Bacterial Components in Inflammation

    Författare :Thorgerdur Sigurdardottir; Anestesiologi och intensivvård; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; sepsis; LL-37; M1 protein; S. pyogenes; Toll-like receptors; phenylephrine; nitr1c oxide; antimicrobial peptides;

    Sammanfattning : Antimicrobial peptides are evolutionally ancient parts of the innate immune system and their primary role is to protect us from infections. The human cathelicidin-derived antimicrobial peptide, LL-37, not only possesses broad spectrum antimicrobial activities but is also able to bind and neutralize bacterial lipopolysaccharide (LPS), an important trigger of the widespread inflammatory response contributing to septic shock.
