1. Temporal monitoring of intracellular Ca2+ signaling and origins of Ca2+ oscillations
Sammanfattning : This thesis examined parameters influencing stimulated cytoplasmic free Ca2+ concentration ([Ca2+]i) oscillations in hepatocytes and pancreatic beta-cells. Hepatic glucose output is regulated in part by hormones such as vasopressin that act through [Ca 2+]i oscillations. LÄS MER
2. Effects of imidazoline compounds on intracellular CA2+ and apoptosis
Sammanfattning : Cytoplasmic free Ca2+ concentration ([Ca2+]i) plays a pivotal role in the regulation of insulin release from the pancreatic beta-cell. [Ca2+]i is also important in the control of apoptosis. LÄS MER
3. Studies on molecular mechanisms in calcium signaling and cellular energy consumption
Sammanfattning : Ion signaling plays fundamental role in cell survival. Na+ and Ca2+ are critical players in ion signaling. Cells spend the major amount of energy to maintain and regulate Na+ and Ca2+ gradients across the cell membrane. Any disruption in cellular energy consumption by plasma membrane ATPases affects ion signaling and vice versa. LÄS MER
4. Metabolic signals in the regulation of insulin release
Sammanfattning : In the present thesis, an attempt was made to elucidate the role of some of the glucose-induced intracellular metabolic effector signals in insulin secretion and to develop new applications that will enable further investigations of oscillatory signalling and secretion from pancreatic ß-cells. The sequence of early metabolic and ionic events was examined in clonal pancreatic insulin secreting cells (HIT) in response to glucose stimulation. LÄS MER
5. Parathyroid hormone-related protein (PTHrP), calcium and human osteoblast-like cells
Sammanfattning : Parathyroid hormone-related protein (PTHrP) may cause humoral hypercalcernia of malignancy (HHM) when overproduced by malignant tumors, but is also produced in lower amounts under normophysiological conditions in practically all cell types, including osteoblasts. One of the aims of the present study was to investigate interactions between PTHrP and osteoblasts, to elucidate bone-regulating functions of PTHrP. LÄS MER
