  1. 1. Functional features of human cytochrome P450 1A2 : with special focus on caffeine and melatonin metabolism

    Författare :Anna Nordmark; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Drug metabolism; CYP1A2; caffeine; melatonin; liver slices; in vitro methods;

    Sammanfattning : CYP1A2 is one of the major cytochrome P450 enzymes in the liver and metabolises drugs such as caffeine, clozapine, tacrine, and theophylline. CYP1A2 also has a role in chemical carcinogenesis due to its activation of procarcinogens, such as heterocyclic amines and arylamines. Caffeine is extensively used to assess CYP1A2 activity. LÄS MER

  2. 2. Metabolism of melatonin : with focus on the influence of cytochrome P4501A2

    Författare :Carina Ursing; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Melatonin; cytochrome P450 1A2 ; fluvoxamine; caffeine; tobacco smoke.;

    Sammanfattning : Melatonin (MT) regulates circadian rhythms. It is produced by pinealocytes. Their function is regulated by light. MT enters the blood stream by passive diffusion. LÄS MER

  3. 3. Consequences of CYP2D6 polymorphism for the disposition and dynamics of tolterodine : a novel drug in the treatment of urinary bladder overactivity

    Författare :Niclas Brynne; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :active metabolite; CYP2D6; CYP3A4; disposition; drug interaction; fluoxetine; ketoconazole; metabolic ratio; pharmacokinetics; tolterodine;

    Sammanfattning : The pharmacokinetics and pharmacological effects of tolterodine were studied in man following administration of increasing oral and intravenous single-doses. The influence of metabolic phenotype in extensive and poor metabolizers of debrisoquine was determined. LÄS MER

  4. 4. Toxicity of smokeless tobacco in human oral epithelium with emphasis on carcinogen metabolism and regulation of programmed cell death

    Författare :Martin Vondracek; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The oral mucosa is globally a common site for cancer development. Primary risk factors include tobacco smoking and alcohol consumption whereas the contribution from usage of smokeless tobacco remains debated. LÄS MER

  5. 5. Importance of pharmacogenetic and environmental factors for variation in caffeine disposition : with special emphasis on CYP1A2, CYP2A6, NAT2 and XO

    Författare :Natasa Djordjevic; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Inter-individual variation in response to drugs conveys considerable risk of either therapy failure or drug toxicity. Caffeine, as one of the most frequently used psychoactive substances in the world, is not an exception, and both lack of effects and adverse reactions have been observed after usual doses. LÄS MER
