Sammanfattning : During the last decade it has been shown that nitric oxide (NO) and carbon monoxide (CO) are produced in our own body, and that they constitute a novel class of messenger molecules. In 1992 a constitutive NO-producing NO-synthase (cNOS) was found in the islets of Langerhans, and in 1997 we could show for the first time that the islets also contained a CO-producing constitutive heme oxygenase (HO-2). 2. Islet constitutive nitric oxide synthase and nitric oxide production. Modulatory effects on insulin and glucagon secretion
Sammanfattning : The aim of this thesis was to elucidate the role of nitric oxide (NO) in the transduction signalling of insulin and glucagon release by stimulating or inhibiting islet endogenous NO-production as well as by using NO donors. Insulin release stimulated by L-arginine, the substrate for constitutive NO-synthase (cNOS)-catalyzed NO-production, was increased by the selective NOS-inhibitor NG-nitro-L-arginine methyl ester (L-NAME) and suppressed by the intracellular NO donor hydroxylamine. 3. Pharmacological control of ciliary activity in the young sea urchin larva : cholinergic and monoaminergic effects and the role of calcium and cyclic nucleotides
Sammanfattning : The ciliary activity of the young (prenervous) sea urchin larva, reflected by its swimming intensity, is measured according to a simple method. 5. Transport and release of insulin granules during biphasic insulin secretion
Sammanfattning : Glucose-evoked insulin secretion exhibits two phases: The first phase represents exocytosis of insulin granules situated at the plasma membrane, whereas the second phase reflects insulin release from granules continuously recruited to the plasma membrane. This thesis investigates different aspects of phasic insulin secretion, with the specific aims to elucidate the possible contribution of insulin granule transport, as well as to study the influence of the nucleotides cAMP and NADPH to this process.
