  1. 1. Oxidative damage and the DNA glycosylase MutYH

    Författare :Kristina Jansson; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Oxidative damage; 8-oxo-7; 8-dihydro-2′-deoxyguanosine; Base excision repair; DNA glycosylase; Mismatch repair; DNA damage checkpoint; 9-1-1 complex; Glutathione-S-transferases; Colon cancer.;

    Sammanfattning : The DNA glycosylase MutYH is highly conserved throughout evolution, and homologs are found in most eukaryotes and prokaryotes examined. MutYH functions as a base excision repair DNA glycosylase that excises adenines misincorporated opposite 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodG), one of the most stable products of oxidative DNA damage. 2. 2. Ribonucleotide reductase and DNA damage

    Författare :Pelle Håkansson; Lars Thelander; Claes Gustafsson; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ribonucleotide reductase; DNA damage; dNTP pools; Medical cell biology; Medicinsk cellbiologi;

    Sammanfattning : A prerequisite for a multicellular organism to survive is the ability to correctly replicate and repair DNA while minimizing the number of heritable mutations. To achieve this, cells need a balanced supply of deoxyribonucleoside triphosphates (dNTPs), the precursors for DNA synthesis. LÄS MER

  3. 3. Induction and repair of clustered DNA damage sites after exposure to ionizing radiation

    Författare :Andris Abramenkovs; Bo Stenerlöw; Peter O'Neill; Uppsala universitet; []
    Nyckelord :NHEJ; DSB repair; clustered DNA damage; DNA repair; DNA-PKcs; HSCS; Ra-223; ionizing radiation; Medicinsk vetenskap; Medical Science;

    Sammanfattning : The mechanisms that maintain genomic stability safeguard cells from constant DNA damage produced by endogenous and external stressors. Therefore, this thesis aimed to specifically address questions regarding the requirement and involvement of DNA repair proteins in the repair of various types of radiation-induced DNA damage. 4. 4. Studies of DNA repair strategies in response to complex DNA damages

    Författare :Ainars Bajinskis; Mats Harms-Ringdahl; Bo Stenerlöw; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; ionizing radiation; complex DNA damage; indirect effect of radiation; dimethyl sulfoxide; molekylärgenetik; Molecular Genetics;

    Sammanfattning : The main aim of this thesis was to study the role of the indirect actions of γ-rays and α-particles on the complexity of primary DNA damages and the repair fidelity of major DNA repair pathways: non-homologous end joining (NHEJ), homologous recombination repair (HRR) and base excision repair (BER). The complexity of radiation-induced damages increases and the proximity between damages decreases with increasing LET due to formation of ionization clusters along the particle track. LÄS MER

  5. 5. Formation and repair of complex DNA damage induced by ionizing radiation

    Författare :Karin Magnander; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ionizing radiation; clustered damage; chromatin structure; DSB;

    Sammanfattning : DNA is the critical target when cells are exposed to ionizing radiation, a potent stressor with capacity to produce complex DNA damages, thereby increasing the risk of cancer. DNA and associated histones form chromatin, which is an effective protection against ionizing radiation. LÄS MER
