1. Alpha-1-microglobulin, heme-binding protein, reductase and antioxidant
Sammanfattning : This thesis describes structural and functional studies of alpha-1-microglobulin (a1m), a 26 kDa plasma and tissue protein that is evolutionarily well-conserved and belongs to the lipocalin superfamily. The lipocalins are polypeptides of 160-190-amino acids that are folded into an 8-stranded beta-barrel forming a pocket with a hydrophobic interior. LÄS MER
2. Structure and Interactions of the Extracellular Matrix Protein PRELP
Sammanfattning : This thesis describes the structure and interactions of a novel extracellular matrix protein - PRELP (proline arginine-rich end leucine-rich repeat protein). The amino acid sequence of PRELP showed that it belonged to the leucine-rich repeat (LRR) protein family in connective tissues. LÄS MER
3. Proteins of the Inter-α-inhibitor Family : Biosynthesis, Plasma Clearance and Interaction with Extracellular Matrix Components
Sammanfattning : Bikunin, a chondroitin sulfate-containing protein of 25 kDa, has protease inhibitory activity and occurs in the plasma in free and complexed form. In inter-α-inhibitor (IαI) and pre-a-inhibitor (PαI) it is covalently linked through its chondroitin sulfate (CS) chain to two or one other polypeptide of about 80 kDa – heavy chains 1 and 2 (H1, H2) and heavy chain 3 (H3) – respectively. 4. The role of leucine-rich repeat glycoproteins/proteoglycans in the assembly of collagen matrices
Sammanfattning : The properties of different connective tissues depend on the synthesis and assembly of macromolecular structures composed of collagen and other extracellular matrix components. Mechanisms modulating these events play important roles in the formation of a functional tissue. LÄS MER
5. Chondrocyte-matrix interactions: with emphasis on the collagen binding integrin a10ß1
Sammanfattning : The work in this thesis has been focused on chondrocyte integrins and their interaction with the matrix proteins chondroadherin (CHAD) and collagen type II (CII). Affinity purification of CHAD-binding proteins from chondrocyte lysate, followed by immunoprecipitation of EDTA-eluted material identified alpha2beta1 as a CHAD-binding integrin. LÄS MER
