  1. 1. Optimization of Colistin Dosage in the Treatment of Multiresistant Gram-negative Infections

    Författare :Matti Karvanen; Otto Cars; Lena E Friberg; Vincent H Tam; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Colistin; CMS; Pharmacology; Pharmacokinetics; PKPD; Antibiotics; Combination therapy; Pharmacometrics; Dosing regimens; Antibiotic resistance; Infectious Diseases; Infektionssjukdomar;

    Sammanfattning : As multidrug resistance in Gram-negative bacilli increases, the old antibiotic colistin has rapidly gained attention as one of few last line treatment options in the form of colistin methanesulfonate (CMS), which is hydrolyzed to colistin both in vitro and in vivo. There is a dearth of knowledge on fundamental aspects of colistin, including pharmacokinetics and optimal dosing regimens. LÄS MER

  2. 2. Physiologically Based Pharmacometric Models for Colistin and the Immune Response to Bacterial Infection

    Författare :Salim Bouchene; Mats O. Karlsson; Wilhelm Huisinga; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; PBPK model; endotoxin; colistin; WBPBPK-PD; CMS; inflammation; tissue distribution; Kp; predictions in tissue; interspecies scaling;

    Sammanfattning : Antibiotic treatment failure might be due to bacterial resistance or suboptimal exposure at target site and there is a lack of knowledge on the interaction between antimicrobial pharmacodynamics (PD) and the immune response to bacterial infections. Therefore, it is crucial to develop tools to increase the understanding of drug disposition to better evaluate antibiotic candidates in drug development and to elucidate the role of the immune system in bacterial infections. 3. 3. Pharmacokinetic and Pharmacodynamic Modeling of Antibiotics and Bacterial Drug Resistance

    Författare :Ami Fazlin Syed Mohamed; Lena Friberg; Teresa Dalla Costa; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacometrics; pharmacokinetics; pharmacodynamics; modeling; antibiotics; resistance; combination; Gram-negative bacteria; gentamicin; colistin; meropenem; newborn infants; critically ill patients; Infectious Diseases; Infektionssjukdomar; Pharmaceutical Science; Farmaceutisk vetenskap;

    Sammanfattning : Exposure to antibiotics is an important factor influencing the development of bacterial resistance.  In an era where very few new antibiotics are being developed, a strategy for the development of optimal dosing regimen and combination treatment that reduces the rate of resistance development and overcome existing resistance is of utmost importance. 4. 4. Study Design and Dose Regimen Evaluation of Antibiotics based on Pharmacokinetic and Pharmacodynamic Modelling

    Författare :Anders Kristoffersson; Lena Friberg; Mats Karlsson; Pernilla Lagerbäck; Otto Cars; Stephan Schmidt; Uppsala universitet; []
    Nyckelord :pharmacometric; optimal design; pharmacokinetics; pharmacodynamics; PKPD; resistance; antibiotics; modeling; time-kill curve; colistin; meropenem; ciprofloxacin; non-linear mixed effects models; bayesian; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : Current excessive use and abuse of antibiotics has resulted in increasing bacterial resistance to common treatment options which is threatening to deprive us of a pillar of modern medicine. In this work methods to optimize the use of existing antibiotics and to help development of new antibiotics were developed and applied. LÄS MER

  5. 5. The role of wastewater in surveillance and emergence of antibiotic resistant bacteria

    Författare :Marion Hutinel; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Microbiology; Bacteriology; Antibiotic resistance; Surveillance; Horizontal gene transfer; HGT; Conjugation; Plasmids; Epidemiology; CPE; E. coli; Carbapenamase; Linezolid; Colistin; Selection; sul4; OXA-48; gar; NDM; mcr; wastewater; sewage; sewer;

    Sammanfattning : As antibiotic resistance spreads among bacterial pathogens, it reduces treatment options and increases treatment failures of infectious diseases. Strategies employed to reduce this spread or adapt to its consequences need to be based on reliable surveillance data which is lacking in many countries, often due to limited resources. LÄS MER
