1. Cis-regulatory variation and divergence in Capsella
Sammanfattning : Cis-regulatory changes in e.g. promoters or enhancers that affect the expression of a linked focal gene have long been thought to be important for adaptation. In this thesis, I investigate the selective importance and genomic correlates of cis-regulatory variation and divergence in the genus Capsella, using massively parallel sequencing data. LÄS MER
2. Advances in studying the role of genetic divergence and recombination in adaptation in non-model species
Sammanfattning : Understanding the role of genetic divergence and recombination in adaptation is crucial to understanding the evolutionary potential of species since they can directly affect the levels of genetic variation present within populations or species. Genetic variation in the functional parts of the genome such as exons or regulatory regions is the raw material for evolution, because natural selection can only operate on phenotypic variation already present in the population. 3. Genetic and Genomic Analysis of DNA Sequence Variation
Sammanfattning : The studies in this thesis describe the application of genotyping and allele specific expression analysis to genetic studies. The role of the gene NPC1 in Triglyceride metabolism was explored in mouse models and in humans on the population level in study I. LÄS MER
4. From Single Gene to Whole Genome Studies of Human Transcription Regulation
Sammanfattning : Transcriptional regulation largely determines which proteins and the protein levels that are found in a cell, and this is crucial in development, differentiation and responses to environmental stimuli. The major effectors of transcriptional regulation are a group of proteins known as transcription factors, which importance is supported by their frequent involvement in mendelian and complex diseases. 5. Elucidating the principles of gene regulation in the human genome
Sammanfattning : Gene regulation is largely controlled by two groups of cis-regulatory elements: (i) promoters that initiate stable mRNAs and (ii) enhancers that generate bidirectionally transcribed unstable enhancer RNAs (eRNAs). While promoters tend to be ubiquitously expressed, enhancers are transcribed in a strongly cell-type specific manner. LÄS MER
