  1. 1. Cell Replacement Therapy for Parkinson's Disease: Potential for Circuitry Repair

    Författare :Tiago Cardoso; Utvecklings- och regenerativ neurobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Rabies-based tracing; Parkinson s disease cell therapy; Cell Replacement Therapy; Human embryonic stem cells; Dopamine neuron; Synaptic integration; circuitry repair;

    Sammanfattning : The derivation of dopamine neurons from human embryonic stem cells (hESCs) now offers a promising alternative to fetal tissue for cell replacement therapy (CRT) in Parkinson´s disease (PD). Using the appropriate chemical cues in vitro, hESCs can be patterned towards bona-fide ventral midbrain (VM) DA neurons that survive, reinnervate, release DA and provide functional recovery when transplanted into rodent and non-human primate models of PD. 2. 2. Neural stem cell engraftment : functional interactions, brain repair and gap junctions

    Författare :Johan Jäderstad; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Neural stem cell (NSC) engraftment therapies show increasing promise as potential cures for several of the diseases of the central nervous system. Traditionally, functional improvements after NSC engraftment have been attributed to replacement of lost neurons and astrocytes, remyelination and support of adjacent cells at risk. LÄS MER

  3. 3. Gene therapy by enzyme replacement for Parkinson’s disease Optimization of continuous DOPA delivery and development of a candidate vector for clinical application

    Författare :Tomas Björklund; Brain Repair and Imaging in Neural Systems (BRAINS); []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; serotonin; Parkinson’s disease; L-DOPA; dyskinesia; GTP cyclohydrolase 1; tyrosine hydroxylase; dopamine; gene therapy; enzyme replacement; gene regulation; aromatic L-amino acid decarboxylase;

    Sammanfattning : The introduction of L-DOPA pharmacotherapy revolutionized the treatment of Parkinson’s disease close to fifty years ago. Before the discovery of dopamine as a neurotransmitter and its involvement in the disease, these patients had very little aid from medication and were bound to a life without movement. LÄS MER

  4. 4. Cell and Gene Therapy for Parkinson's Disease: Therapeutic Effect and Modulation of Dyskinesias in the 6-OHDA Rat Model

    Författare :Thomas Carlsson; Brain Repair and Imaging in Neural Systems (BRAINS); []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neuropsykologi; Pharmacological sciences; pharmacognosy; pharmacy; neurofysiologi; Medicin människa och djur ; Neurology; neuropsychology; neurophysiology; Neurologi; Medicine human and vertebrates ; Parkinson?s disease; Serotonin; Tyrosine hydroxylase; Ventral mesenchephalon; Motor behavior; L-DOPA; GTP cyclohydrolase 1; Gene therapy; Dyskinesias; Cell transplantation; Dopamine; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi;

    Sammanfattning : The main pathological feature in Parkinson's disease is the progressive loss of dopamine neurons in the midbrain, which in turn leads to the appearance of motor deficits such as akinesia/bradykinesia (loss/slowness of movements), rigidity, postural imbalance and tremor. To this day, there is no cure for the disease, but there are medications to relieve the symptoms. LÄS MER

  5. 5. Synaptic Integration of Hippocampal Neurons Generated in the Adult Brain: Influence of a Pathological Environment

    Författare :Katherine Jakubs; Lund Neurologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neuropsykologi; neurofysiologi; Neurologi; synaptic plasticity; GABA; glutamate; GFP; Neurology; neuropsychology; neurophysiology; hippocampus; dentate gyrus; neurogenesis; LPS; inflammation; epilepsy; SE; patch-clamp;

    Sammanfattning : The adult brain constantly produces new neurons from endogenous neural progenitor cells located in at least two regions, the subgranular zone (SGZ) in dentate gyrus of the hippocampus, and the subventricular zone (SVZ). Major efforts are underway to discover the functional significance of new neurons in the adult brain. LÄS MER
