  1. 21. Linker histone H1 and androgen receptor : two different players in the chromatin orchestra

    Författare :Christine Öberg; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The linker histone H1 and the androgen receptor are two different players in the chromatin orchestra. The linker histone H1, one of the most abundant proteins in the nucleus, is located at the surface of the nucleosome but despite many important functions reported for this protein it is not as well studied as the core histones. 2. 22. Beyond transcription : a post-transcriptional role of 3D chromatin crosstalk in oncogene regulation

    Författare :Johanna Vestlund; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : This thesis explores how stochastic chromatin fibre interactions, chromatin organization in the 3D nuclear architecture, and environmental signals collaborate to regulate MYC oncogene expression in human colon cancer cells. In Paper I, we employ the ultra-sensitive Nodewalk technique to uncover the dynamic and stochastic nature of chromatin networks impinging on MYC. LÄS MER

  3. 23. Crosstalk between environmental signals and 3d genome organization in the regulation of gene expression

    Författare :Anna Lewandowska Ronnegren; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The thesis explores the connection between environmental stimuli and gene expression regulated by the spatial changes in genome organization. In Paper I, by applying state of the art Circular Chromosome Conformation Capture assay (4C) and Chromatin in situ Proximity (ChrISP) techniques, we show that transcriptionally active circadian genes meet in space with repressed lamina-associated domains (LADs), and that these interactions are under the control of the circadian clock. 4. 24. Studies of the chromatin form of yeast Mediator and the function of its tail module

    Författare :Miriam Nilsson; Stefan Björklund; Claes Gustafsson; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Mediator complex; Transcriptional regulation; yeast; tail module; transcription; medicinsk biokemi; Medical Biochemistry; molekylärbiologi; Molecular Biology;

    Sammanfattning : Mediator is a large, multifunctional complex that is conserved in eukaryotes. It was first discovered in Saccharomyces cerevisiaeas required for transcriptional activators to function in a reconstituted in vitro system. Mediator is also important for stimulation of basal, unregulated transcription, and transcriptional repression. LÄS MER

  5. 25. Novel methods for mapping genome organization and genome fragility in the 3D space of the nucleus

    Författare :Tomasz Kallas; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : For over a century, scientists have been trying to understand how the DNA molecule that is so tightly packed in the micro space of the nucleus sustains critical cellular processes such as transcription, replication or the maintenance of genetic information. Despite the huge effort of researchers around the world, we know relatively little. LÄS MER
