  1. 11. Exploring the Interplay of Lipids and Membrane Proteins

    Författare :Candan Ariöz; Åke Wieslander; Daniel O. Daley; Malcolm East; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Membrane lipids; membrane proteins; anionic lipids; membrane remodeling; intracellular vesicles; model membrane systems; glycosyltransferase; Escherichia coli; lipid composition; fatty acid modification; membrane curvature; bacterial homeoviscous adaptation; Biochemistry; biokemi;

    Sammanfattning : The interplay between lipids and membrane proteins is known to affect membrane protein topology and thus have significant effect (control) on their functions. In this PhD thesis, the influence of lipids on the membrane protein function was studied using three different membrane protein models. LÄS MER

  2. 12. Thiopurine S-methyltransferase - characterization of variants and ligand binding

    Författare :Annica Blissing; Lars-Göran Mårtensson; Tomas Nyman; Linköpings universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; Thiopurine S-methyltransferase TPMT ; protein stability; enzyme kinetics; ligand binding; isothermal titration calorimetry ITC ; 8-anilinonaphthalene-1-sulfonic acid ANS ;

    Sammanfattning : Thiopurine S-methyltransferase (TPMT) belongs to the Class I S-adenosylmethionine-dependent methyltransferase (SAM-MT) super family of structurally related proteins. Common to the members of this large protein family is the catalysis of methylation reactions using S-adenosylmethionine (SAM) as a methyl group donor, although SAM-MTs act on a wide range of different substrates and carry out numerous biologically important functions. 3. 13. Generation and exploitation of proton motive force: Biochemical and structural analysis of three bacterial integral membrane proteins

    Författare :Pontus Emanuel Gourdon; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; X-ray crystallography; AcrB; structure; proteorhodopsin; multi-drug resistance; transhydrogenase; membrane proteins; proton motive force;

    Sammanfattning : Proton motive force, a necessity for all living cells, is generated and exploited by asignificant number of membrane-bound enzymes and transporters. The focus of thisthesis was to understand the physiological relevance, structure and mechanism of threesuch bacterial enzymes: proteorhodopsin; transhydrogenase; and AcrB. LÄS MER

  4. 14. On the crystallization of membrane proteins in lipidic sponge and cubic phases

    Författare :Pia Jeanette Hindrichsen; Chalmers tekniska högskola; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; monoolein MO ; selachyl alcohol; cubic phase; sponge phase; small angle X-ray scattering; protein crystallization; phytantriol PT ;

    Sammanfattning : Membrane proteins are involved in many important biological processes and in order to understand their mechanism, their three-dimensional structures need to be elucidated to high resolution by for example X-ray crystallography. However, there is only limited structural knowledge for membrane proteins which is partly explained by the difficulties in obtaining well-diffracting crystals. LÄS MER

  5. 15. Caspase activation in human neuroblastoma cells: mechanisms and spatiotemporal aspects

    Författare :Elena V. Ivanova; Kerstin Iverfeldt; Boris Zhivotovsky; Stockholms universitet; []

    Sammanfattning : Apoptosis is one of the modes of programmed cell death, in which several members of the caspase family of proteases play the central role. However, activation of apoptotic caspases does not necessarily lead to cell death. LÄS MER
