  1. 1. Prognostic Biomarkers and Target Proteins for Treatment of High-grade Gliomas

    Författare :Linda Sooman; Simon Ekman; Johan Lennartsson; Joachim Gullbo; Michael Bergqvist; Erik Blomquist; Stefan Bergström; Beatrice Melin; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; High-grade glioma; prognostic biomarkers; combination chemotherapy; DNA methylation; FGF2; p38 MAPK; PTPN6; camptothecin; imatinib; vandetanib; EGFR; RAC1; Notch; Medical Science; Medicinsk vetenskap; Oncology; Onkologi;

    Sammanfattning : The survival for high-grade glioma patients is poor and the treatment may cause severe side effects. A common obstacle in the treatment is chemoresistance. To improve the quality of life and prolong survival for these patients prognostic biomarkers and new approaches for chemotherapy are needed. LÄS MER

  2. 2. Application of new methodology for preclinical development of anticancer drugs : With special focus on camptothecin derivatives and the cyanoguanidine CHS 828

    Författare :Elin Jonsson; Uppsala universitet; []
    Nyckelord :Medical sciences; drug development; hollow-fiber model; FMCA; human tumour cell; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; klinisk farmakologi; Clinical Pharmacology;

    Sammanfattning : This work describes the development and evaluation of new methods for preclinical anticancer drug development. The methods were specifically applied to study camptothecin analogues and the new compound CHS 828 (N-(4-chlorophenoxyhexyl)-N'-cyano-N"-4-pyridylguanidine) currently in early clinical trials. 3. 3. Targeting of the Myc pathway as a novel approach for cancer therapy

    Författare :Hao Mo; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :oncogene; tumor suppressor gene; Myc; cellular screening; small molecules; MYRAs; NSC308848; camptothecin; paclitaxel; apoptosis; transcription; transformation.;

    Sammanfattning : As transcription factors, Myc-family proteins function upstream of signaling pathways and regulate cell proliferation, differentiation, angiogenesis, apoptosis and transformation. Precisely regulated Myc expression is critical for cellular physiology, while perturbations of Myc expression have detrimental consequences to the organism. LÄS MER

  4. 4. Roles of Myc and Mad in cell cycle and apoptosis

    Författare :Ami Albihn; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Myc; Mad; cell cycle; apoptosis; differentiation; tumorigenesis; chemotherapy;

    Sammanfattning : The Myc network proteins are key mediators in regulation of cell growth, differentiation and apoptosis. They are basic region hehx-loop-helix/leucine zipper (bHLH/Zip) transcription factors that require hetero-dimerization with Max for specific DNA binding Mad family members are expressed primarily in differentiated tissues where they recruit histone deacetylase complexes via the mSin3 interaction domain (SID) to repress transcription of target genes and prevent cell growth In contrast, members of the Myc family activate target gene transcription by recruitment of histone acetyltransferases to their transcriptional activation domain (TAD), inducing proliferation and S phase entry.
