  1. 11. Radiolabelled Oligonucleotides for Evaluation of in vivo Hybridisation Utilising PET Methodology

    Författare :Gábor Lendvai; Mats Bergström; Sergio Estrada; Barbro Eriksson; Raymond Josephsson; Bertrand Tavitian; Uppsala universitet; []
    Nyckelord :Molecular medicine; Gene expression; Antisense oligonucleotides; Positron emission tomography; In vivo hybridisation; In vivo biodistribution; Chromogranin-A; 68Ga; RT-PCR; Scavenger receptors; Molekylärmedicin;

    Sammanfattning : Antisense oligonucleotides (ODN) may interfere in gene expression on the basis of hybridising to its complementary messenger RNA (mRNA) sequence in the cell thereby preventing the synthesis of the peptide. Therefore, these ODNs may be potential drugs to treat human diseases by “knocking down” the expression of responsible genes or correcting the maturation process of mRNA in the field called antisense therapy. 2. 12. Influence of extrinsic factors on bone scintigraphy an experimental and clinical study

    Författare :Marika Cronhjort; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Bone scintigraphy; bisphosphonates; biodistribution; animal;

    Sammanfattning : Bone scintigraphy takes advantage of the strong affinity of technetiated diphosphonates to bone. These compounds accumulate particularly at sites of new bone formation which occurs as a response to various conditions affecting bone. LÄS MER

  3. 13. Development of ADAPT-based tracers for radionuclide molecular imaging of cancer

    Författare :Javad Garousi; Vladimir Tolmachev; Tony Lahoutte; Uppsala universitet; []
    Nyckelord :ADAPT; scaffold protein; albumin-binding domain ABD ; Affinity protein; HER2; radionuclide molecular imaging;

    Sammanfattning : ABD-Derived Affinity Proteins (ADAPTs) is a novel class of small engineered scaffold proteins based on albumin-binding domain (ABD) of streptococcal protein G. High affinity ADAPT  binders against various therapeutic targets can be selected. LÄS MER

  4. 14. Multicomponent Reactions in 11C/12C Chemistry : – Targeting the Angiotensin II Subtype 2 Receptor

    Författare :Marc Stevens; Luke Odell; Mats Larhed; Gunnar Antoni; Sergio Estrada; Ola Åberg; Neil Vasdev; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; carbonylation; positron emission tomography; multicomponent reactions; AT2R; 3; 4-dihydroquinazolinone; sulfonyl azides; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : Section 1 of this thesis contains an introduction to method development in organic synthesis, multicomponent reactions, sulfonyl azides, tracer development in 11C chemistry and the biological target.Section 2 describes the use of sulfonyl azides in carbonylative chemistry. Paper I covers development of a diazotransfer protocol. LÄS MER

  5. 15. Engineering of Affibody molecules for Radionuclide Molecular Imaging and Intracellular Targeting

    Författare :Camilla Hofström; Torbjörn Gräslund; Serge Muyldermans; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Affibody molecules; affinity proteins; radionuclide molecular imaging; intracellular targeting; EGFR; HER2; IGF-1R; SRA - Molecular Bioscience; SRA - Molekylär biovetenskap;

    Sammanfattning : Affibody molecules are small (7 kDa) affinity proteins of non-immunoglobulin origin that have been generated to specifically interact with a large number of clinically important molecular targets.In this thesis, Affibody molecules have been employed as tracers for radionuclide molecular imaging of HER2- and IGF-1R-expressing tumors, paper I-IV, and for surface knock-down of EGFR, paper V.
