1. In vivo Pharmacokinetic Interactions of Finasteride and Identification of Novel Metabolites
Sammanfattning : The general aim of this thesis was to improve the understanding of the in vivo pharmacokinetics and, in particular, the metabolism of finasteride, a 5α-reductase inhibitor used in the treatment of enlarged prostate glands and male pattern baldness. CYP3A4 has been identified as the major enzyme involved in the sequential metabolism of finasteride to ω-OH finasteride (M1) and ω-COOH finasteride (M3). LÄS MER
2. The Hepatobiliary Transport of Rosuvastatin In Vivo
Sammanfattning : In vivo studies of hepatobiliary disposition are challenging. The hepatobiliary system is complex, as its physiological localization, complex cellular structure with numerous transporters and enzymes, and the interindividual variability in protein expression and biliary flow will all affect the in vivo disposition of a drug under investigation. 3. In vivo Pharmacokinetics of Two New Thrombin Inhibitor Prodrugs : Emphasis on Intestinal and Hepatobiliary Disposition and the Influence of Interacting Drugs
Sammanfattning : Biliary excretion is an important elimination route for many drugs and metabolites. For such compounds, it is important to know the extent of excretion and drug exposure in the bile, e.g., for the risk assessment of drug interactions, liver toxicity and the effects of genetic variants. 4. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury
Sammanfattning : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 5. Image-based dynamic liver testing : studies of segmental hepatic parenchymal function and biliary flow using dynamic 99Tcm-HIDA SPECT
Sammanfattning : The aim of the study was to create a dynamic liver function test using a tomographic imaging procedure as sampling method. The liver has multiple and complex functions. It is obvious that no single test can give a comprehensive measurement of total liver function. LÄS MER
