  1. 1. In vivo Pharmacokinetics of Two New Thrombin Inhibitor Prodrugs : Emphasis on Intestinal and Hepatobiliary Disposition and the Influence of Interacting Drugs

    Författare :Elin Matsson; Hans Lennernäs; Lars Knutson; Ulf Eriksson; Kim Brouwer; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Direct thrombin inhibitors; prodrugs; ximelagatran; AZD0837; erythromycin; ketoconazole; drug-drug interactions; hepatobiliary transport; biliary clearance; ATP-binding cassette; ABC transporters; solute carriers; SLC transporters; CYP3A4; Biopharmacy; Biofarmaci; Biopharmaceutics; Biofarmaci;

    Sammanfattning : Biliary excretion is an important elimination route for many drugs and metabolites. For such compounds, it is important to know the extent of excretion and drug exposure in the bile, e.g., for the risk assessment of drug interactions, liver toxicity and the effects of genetic variants. 2. 2. In vivo Pharmacokinetic Interactions of Finasteride and Identification of Novel Metabolites

    Författare :Anna Lundahl; Hans Lennernäs; Dennis A Smith; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug-drug interactions; herb-drug interactions; pharmacokinetics; metabolism; CYP3A4; hydroxylation; glucuronidation; UGT enzymes; mass spectrometry; biliary excretion; urinary excretion; finasteride; PHARMACY; FARMACI;

    Sammanfattning : The general aim of this thesis was to improve the understanding of the in vivo pharmacokinetics and, in particular, the metabolism of finasteride, a 5α-reductase inhibitor used in the treatment of enlarged prostate glands and male pattern baldness. CYP3A4 has been identified as the major enzyme involved in the sequential metabolism of finasteride to ω-OH finasteride (M1) and ω-COOH finasteride (M3). LÄS MER

  3. 3. Complicated gallstone disease in Sweden 1988-2006 : a register study

    Författare :Birger Sandzén; Mikael Öman; Erik Nilsson; Roland Andersson; Umeå universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Gallstone disease; acute pancreatitis; acute biliary pancreatitis; common bile duct stones; acute cholecystitits; acute gallbladder disease; cholecystectomy; ERCP; endoscopic papillotomy; choledochotomy; Surgery; Kirurgi; kirurgi; Surgery;

    Sammanfattning : Background The gallstone prevalence in the western world is 10-20%. Most gallstones are silent, but symptoms and complications appear in 20-40%. The incidence of symptom development in patients with silent gallstones is 2-4% per year. LÄS MER

  4. 4. Image-based dynamic liver testing : Studies of segmental hepatic parenchymal function and biliary flow using dynamic 99Tcm-HIDA SPECT

    Författare :Eduard Jonas; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; Dynamic SPECT; Segmental liver function test; Primary sclerosing cholangitis; Bile flow; Bile scintigraphy;

    Sammanfattning : The aim of the study was to create a dynamic liver function test using a tomographic imaging procedure as sampling method. The liver has multiple and complex functions. It is obvious that no single test can give a comprehensive measurement of total liver function. LÄS MER

  5. 5. Aspects of interventional endoscopic treatment of common bile duct stones

    Författare :Fredrik Swahn; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; ERCP;

    Sammanfattning : Background: During the last 30 years, important changes have been introduced in the management of common bile duct stones (CBDS) which can be detected in about 10% of patients undergoing cholecystectomy because of symptomatic gallstone disease. Established minimally-invasive treatment options for CBDS include endoscopic retrograde cholangiopancreatography (ERCP) and laparoscopic common bile duct exploration. LÄS MER
