1. In vivo Pharmacokinetics of Two New Thrombin Inhibitor Prodrugs : Emphasis on Intestinal and Hepatobiliary Disposition and the Influence of Interacting Drugs
Sammanfattning : Biliary excretion is an important elimination route for many drugs and metabolites. For such compounds, it is important to know the extent of excretion and drug exposure in the bile, e.g., for the risk assessment of drug interactions, liver toxicity and the effects of genetic variants. Sammanfattning : The general aim of this thesis was to improve the understanding of the in vivo pharmacokinetics and, in particular, the metabolism of finasteride, a 5α-reductase inhibitor used in the treatment of enlarged prostate glands and male pattern baldness. CYP3A4 has been identified as the major enzyme involved in the sequential metabolism of finasteride to ω-OH finasteride (M1) and ω-COOH finasteride (M3). LÄS MER
Sammanfattning : Background The gallstone prevalence in the western world is 10-20%. Most gallstones are silent, but symptoms and complications appear in 20-40%. The incidence of symptom development in patients with silent gallstones is 2-4% per year. LÄS MER
4. Image-based dynamic liver testing : Studies of segmental hepatic parenchymal function and biliary flow using dynamic 99Tcm-HIDA SPECT
Sammanfattning : The aim of the study was to create a dynamic liver function test using a tomographic imaging procedure as sampling method. The liver has multiple and complex functions. It is obvious that no single test can give a comprehensive measurement of total liver function. LÄS MER
Sammanfattning : Background: During the last 30 years, important changes have been introduced in the management of common bile duct stones (CBDS) which can be detected in about 10% of patients undergoing cholecystectomy because of symptomatic gallstone disease. Established minimally-invasive treatment options for CBDS include endoscopic retrograde cholangiopancreatography (ERCP) and laparoscopic common bile duct exploration. LÄS MER
