1. Amino acids with relevance to health, climate and the environment : Development of mass spectrometric methods
Sammanfattning : Amino acids play vital roles in health, either in their native form or chemically modified. Some studies have linked certain non-proteinogenic amino acids to neurodegenerative diseases, such as in the case of β-methylaminoalanine (BMAA). LÄS MER
2. Studies on oligonucleotide adducts formed from 7,8-dihydroxy-9,10-epoxy-7,8,9,10-tetrahydrobenzo[A]pyrene stereoisomers
Sammanfattning : The widespread environmental pollutant benzo[a]pyrene (BP), is formed from incomplete combustion of fossil fuels and is mutagenic and carcinogenic after metabolic activation to bay-region diol epoxides and subsequent covalent binding to DNA. The metabolic activation pathway to diol epoxides is via BP-7,8-oxide and BP-7,8-dihydrodiol to yield preferentially 7R,8S-dihydroxy-9S,lOR-epoxy-7,8,9,10-tetrahydrobenzo[a]py- rene [(+)-anti-BPDE] and to lesser extents, 7R,8S-dihydroxy-9R,lOS-epoxy-BP [(-)-syn-BPDE], 7S,8R- dihydroxy-9S,lOR-epoxy-BP [(+)-syn-BPDE] and 7S,8R-dihydroxy-9R,lOS-epoxy-BP [(-)-anti-BPDE]. 3. Polycyclic aromatic hydrocarbons : DNA damage and cell signaling
Sammanfattning : Oxidative stress is a threat to our wellbeing. The formation of reactive oxygen species may result in oxidative lesions in DNA and RNA. When challenging A549 cells with 18O-labeled hydrogen peroxide, we found that RNA was between 14-25 times more sensitive to [18O]-8-oxoGuo formation than [18O]-8-oxodGuo formation in DNA. LÄS MER
4. Carcinogenic polycyclic aromatic hydrocarbons : role of human glutathione transferases in conjugate formation and DNA protection
Sammanfattning : Polycyclic aromatic hydrocarbons (PAHs) are widespread mutagenic and carcinogenic environmental pollutants, which requires metabolic activation to electrophilic intermediates and subsequent covalent binding to critical targets in DNA to elicit their biological activity. Bay- and fjord region diol epoxides (DEs) have been identified as the ultimate mutagenic and carcinogenic metabolites of PAHs. LÄS MER
