1. Membrane Interactions of Arginine-Rich Peptides for the Intracellular Delivery of Gene-Targeted Drugs
Sammanfattning : A major obstacle for the therapeutic use of e.g. oligonucleotides is their inherently poor cellular uptake. A recently discovered class of peptides, denoted cell-penetrating peptides, has been shown to traverse cell membranes, even when conjugated to oligonucleotides or even full-length proteins. LÄS MER
2. Aspects of Antisense and Antigene Chemistry of Oligonucleotides Tethered to Intercalators
Sammanfattning : Synthetic and physicochemical studies on appropriately functionalized ODN-conjugates have been performed to evaluate their abilities to act as antisense agents against RNA or as intramolecular DNA cross-linking agents. Intercalating aromatic systems [phenazine (Pnz), dipyridophenazine (DPPZ)] and metallointercalators such as Ru2+(phen)2(DPPZ) and Ru2+(tpy)(DPPZ)L [where L = chemically or photochemically labile ligand, phen = phenanthroline, tpy = terpyridine], which are covalently tethered to the oligo-deoxynucleotides (ODNs), have been chosen for this purpose. 3. Towards smart antisense and antigene therapeutics : design and synthesis of novel oligonucleotide based bioconjugates
Sammanfattning : The use of nucleic acid technology has been of great importance in life science research. In several clinical trials patients with genetic disease are treated with help of innovations based on oligonucleotide therapy. The use of it is unfortunately limited in efficacy due to e.g. LÄS MER
4. Nucleoside phosphoramidates and related compounds : development of synthetic methods based on H-phosphonate chemistry
Sammanfattning : In this thesis, studies directed towards development of new synthetic methods for the preparation of nucleotide analogues of potential medical value as antisense, antigene or antiviral agents, are described. Two methods for synthesis of N3'->P5' and P3'->N5' phosphoramidate analogues with the nitrogen atom in bridging position of the internucleotide linkage was developed. 5. Optimization of the formulation and design of oligonucleotide-based pharmaceuticals for the purpose of gene therapy
Sammanfattning : Oligonucleotides (ONs) are short sequences of nucleic acids which may be used in a therapeutic context to modulate gene expression. According to their target, ONs can be classified into two main classes: antisense ONs which target mRNA and antigene ONs that target chromosomal DNA. LÄS MER
