1. Surface Properties of Antibodies and their Complexes with Antigens Studied by LLPC
Sammanfattning : The main objective of this thesis was to gain further insight into the relationship between the exposed surfaces and the functional properties of antibodies and their complexes with antigens in solution. To study this relationship, a new technique, liquid-liquid partition chromatography (LLPC) in aqueous two-phase systems, was developed further with respect to sensitivity, selectivity and reproducibility. 2. Antigen interaction with B cells in two proliferative disorders : CLL and MGUS
Sammanfattning : The aim of the work presented in this thesis was to elucidate B cell interaction with antigen in the two B cell proliferative disorders chronic lymphocytic leukemia (CLL) and monoclonal gammopathy of undetermined significance (MGUS). In the first part we investigated the antigen specificity of CLL cells and characterized Epstein-Barr virus (EBV)-transformed CLL cell lines with regard to phenotype and genotype. LÄS MER
3. Aβ Conformation Dependent Antibodies and Alzheimer's Disease
Sammanfattning : Soluble intermediates of the amyloid-β (Aβ) aggregation process are suggested to play a central role in the pathogenesis of Alzheimer’s disease (AD) by causing synaptic dysfunction and neuronal loss. In this thesis, soluble Aβ aggregates have been studied with a particular focus on the Aβ protofibril, which has served as the antigen for developing conformation dependent monoclonal antibodies. 4. Antibody-Fragments: Protein Engineering, Process Development, and their Application as Affinity Ligands
Sammanfattning : Monoclonal antibody (mAb) fragments have become the formats of choice for both biopharmaceuticals and ligands in analytical and preparative applications. Their smaller size enables additional advantages over full-length mAbs, such as improved tissue penetration, faster blood clearance for diagnostic applications, improved capacity, and enablement of multivalent formatting. 5. Targeting cytotoxic species in amyloid diseases
Sammanfattning : Amyloid diseases are a world-wide problem causing great human suffer and large economical costs. Although amyloid deposits, a common denominator in all amyloid disorders, are detrimental to the surrounding tissue, there is a poor correlation between total amyloid burden and clinical symptoms.
