11. Human antibody repertoires in normal physiology and in autoimmune disease
Sammanfattning : The immune system has to balance the need for a broad protection from infectious agents against the risk of developing autoimmune disease. The elements of the immune system should provide clues about conditions under which physiological autoreactivity may develop into autoaggressive immunity, if analyzed against the background of the immune system in healthy individuals. 12. Biomarker Discovery in Cancer using Affinity Proteomics - Studies of B-cell lymphomas and Breast Cancer
Sammanfattning : The continual increase in incidence of cancer has created an urgent need for improved methods for diagnosis, prognosis and/or treatment. In this context, the field of biomarker research has expanded immensely during the last few years. This thesis, based on four original papers, revolves around protein biomarker discovery in cancer. 13. Repertoire diversity and maturation of HIV-1 vaccine-induced B cell responses
Sammanfattning : In the work presented in this thesis, I characterized B cell responses in rhesus macaques inoculated with HIV-1 envelope glycoprotein (Env)-based vaccines to define genetic and functional properties of the elicited antibodies for an improved understanding of Env immunogenicity. In Paper I, we investigated antibody heavy chain V gene segment expression in the overall B cell repertoire and in response to vaccination by using 2 approaches, 454-pyrosequencing and single-cell RT-PCR of bulk and sorted memory B cells, respectively. 14. The clinical importance of non-HLA specific antibodies in kidney transplantation
Sammanfattning : The clinical significance of human leukocyte antigen (HLA) antibodies (Abs) for hyperacute, acute and chronic antibody-mediated rejection (AMR) of kidney allografts has been clearly demonstrated. AMR occurs in the absence of donor-reactive HLA Abs. LÄS MER
15. NK cell recognition : adaptability to host factors in normal and diabetic mice
Sammanfattning : The overall aim of this thesis was to understand the rules, which guide NK cell behavior in vivo. NK cells are part of the innate immune system and are important in defense against certain virus infections and tumorigenic cells. They also play a role in rejecting bone marrow grafts. LÄS MER
