  1. 1. Epac2 signaling at the β-cell plasma membrane

    Författare :Ida Alenkvist; Anders Tengholm; Olga Göransson; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; β-cell; insulin secretion; cAMP; Ca2 ; Epac; Rap; exocytosis; sulfonylurea; Medical Cell Biology; Medicinsk cellbiologi;

    Sammanfattning : Secretion of appropriate amounts of insulin from pancreatic β-cells is crucial for glucose homeostasis. The β-cells release insulin in response to glucose and other nutrients, hormones and neurotransmitters, which trigger intracellular signaling cascades, that result in exocytotic fusion of insulin-containing vesicles with the plasma membrane. 2. 2. Oscillatory Signaling and Insulin Secretion from Single ß-cells

    Författare :Olof Idevall Hagren; Anders Tengholm; Erik Gylfe; Leslie Satin; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cAMP; Ca2 ; oscillations; beta-cell; insulin secretion; evanescent wave microscopy; PIP3; PKA; Epac; Medical cell biology; Medicinsk cellbiologi; Medical Cell Biology; Medicinsk cellbiologi;

    Sammanfattning : cAMP and Ca2+ are key regulators of exocytosis in many cells, including insulin-secreting pancreatic β-cells. Glucose-stimulated insulin secretion from β-cells is pulsatile and driven by oscillations of the cytoplasmic Ca2+ concentration ([Ca2+]i), but little is known about the kinetics of cAMP signaling and the mechanisms of cAMP action. 3. 3. Studies on molecular mechanisms in calcium signaling and cellular energy consumption

    Författare :Kalaiselvan Krishnan; Hjalmar Brismar; Anders Tengholm; KTH; []
    Nyckelord :ca2 signaling; ATP; PercevalHR; NKA; diabetes; Angiotensin; Biological Physics; Biologisk fysik;

    Sammanfattning : Ion signaling plays fundamental role in cell survival. Na+ and Ca2+ are critical players in ion signaling. Cells spend the major amount of energy to maintain and regulate Na+ and Ca2+ gradients across the cell membrane. Any disruption in cellular energy consumption by plasma membrane ATPases affects ion signaling and vice versa. LÄS MER

  4. 4. ATP Dynamics in Pancreatic α- and β-cells

    Författare :Jia Li; Anders Tengholm; Erik Gylfe; Hindrik Mulder; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; ATP; Ca2 ; β-cell; α-cell; SERCA; SOCE; muscarinic receptor; insulin secretion; glucagon secretion; Medical Cell Biology; Medicinsk cellbiologi;

    Sammanfattning : Glucose metabolism in pancreatic α- and β-cells is believed to regulate secretion of glucagon and insulin, respectively. In β-cells, ATP links glucose metabolism to electrical activity and insulin secretion. LÄS MER

  5. 5. MEKK-1 and NF-κB Signaling in Pancreatic Islet Cell Death

    Författare :Dariush Mokhtari; Nils Welsh; Michael Welsh; Anders Tengholm; Shanta Persaud; Uppsala universitet; []
    Nyckelord :Cell biology; MEKK-1; JNK; human islets; β-cells; siRNA; apoptosis; cell death; MAPK; nitric oxide; cytokines; streptozotocin; c-Rel; NF-κB; Cellbiologi;

    Sammanfattning : Type 1 diabetes is an autoimmune disease resulting in the selective destruction of the insulin producing β-cells in the pancreas. Pro-inflammatory cytokines and the free radical nitric oxide (NO) have been implicated in mediating the destruction of β-cells, possibly through activation of the mitogen activated protein kinases (MAPKs) JNK, ERK and p38.
