  1. 1. Engineering enzymes towards biotherapeutic applications using ancestral sequence reconstruction

    Författare :Natalie Hendrikse; Per-Olof Syrén; Johan Rockberg; Erik Nordling; Reinhard Sterner; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; enzyme engineering; enzyme evolution; ancestral sequence reconstruction; therapeutic enzymes; terpene cyclases; ammonia-lyases; sulfatases; lysosomal enzymes; Kemi; Chemistry;

    Sammanfattning : Enzymes are versatile biocatalysts that fulfill essential functions in all forms of life and, therefore, play an important role in health and disease. One specific application of enzymes in life science is their use as biopharmaceuticals, which typically benefits from high catalytic activity and stability. LÄS MER

  2. 2. Computational problems in evolution : Multiple alignment, genome rearrangements, and tree reconstruction

    Författare :Isaac Elias; Jens Lagergren; Lior Pachter; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Distance Methods; Tree reconstruction; Sorting by Transpositions; multiple alignment; ancestral sequences; Computer science; Datavetenskap;

    Sammanfattning : Reconstructing the evolutionary history of a set of species is a fundamental problem in biology. This thesis concerns computational problems that arise in different settings and stages of phylogenetic tree reconstruction, but also in other contexts. LÄS MER

  3. 3. The Dawn of a New Age : Interrelationships of Acoela and Nemertodermatida and the Early Evolution of Bilateria

    Författare :Andreas Wallberg; Ulf Jondelius; Mikael Thollesson; Graham Budd; Gonzalo Giribet; Uppsala universitet; []

    Sammanfattning : Deciphering the rapid emergence of bilaterian animals around the time of the Cambrian Explosion and reconstructing the interrelationships of animal groups have long been two of the most elusive problems in Zoology. This thesis concerns the phylogenetic interrelationships within and among Acoela and Nemertodermatida, two groups of small worms that are believed to be basal bilaterians and which may provide important clues for understanding the early evolution of animals. 4. 4. Genomic and evolutionary exploration of Asgard archaea

    Författare :Eva F. Caceres; Thijs J. G. Ettema; Simonetta Gribaldo; Uppsala universitet; []
    Nyckelord :archaea; Asgard; eukaryogenesis; metagenomics; genome binning; phylogenetics; phylogenomics; comparative genomics; gene tree-species tree reconciliation; ancestral reconstruction; long-read metagenomics;

    Sammanfattning : Current evolutionary theories postulate that eukaryotes emerged from the symbiosis of an archaeal host with, at least, one bacterial symbiont. However, our limited grasp of microbial diversity hampers insights into the features of the prokaryotic ancestors of eukaryotes. LÄS MER

  5. 5. In silico protein design for the enhancement of protein stability and function

    Författare :David A. Hueting; Per-Olof Syrén; Hjalmar Brismar; Zbynek Prokop; KTH; []

    Sammanfattning : Enzymes are natures catalysts that increase the rate of a chemical reaction. The increased rate of a reaction is required to be able to sustain life. Despite the huge impact of enzymes, they are not perfect catalysts. LÄS MER
