6. A beta-phosphoglucomutase in carbohydrate metabolism of Lactococcus lactis
Sammanfattning : Phosphoglucomutase (PGM) catalyzes the reversible conversion of glucose 1-phosphate to glucose 6-phosphate. Two distinct forms of PGM have been purifed and charactierized in Lactococcus lactis growing on maltose: one specific for the alpha-isomer, and the other specific for the beta-isomer of glucose 1-phosphate. 7. Ferrochelatase and Magnesiumchelatase: Metal chelation studied with mutants
Sammanfattning : Magnesium chelatase and ferrochelatase are two very important enzymes, which are involved in chlorophyll and heme biosynthesis, respectively. They both use protoporphyrin IX as substrate but magnesium chelatase inserts a magnesium ion whereas ferrochelatase inserts a ferrous ion. These two enzymes have been studied in this thesis. LÄS MER
8. Metabolic analysis of shikimic acid producing Escherichia coli
Sammanfattning : Shikimic acid production using recombinant E.coli has been developed during the past 15 years due to the increased demand of this product, primarily as a starting material for the anti-influensa medicine Tamiflu?. A detailed metabolic analysis of a shikimic acid producing E. LÄS MER
9. Peptide-tagged proteins in aqueous two-phase systems
Sammanfattning : This thesis deals with proteins containing peptide tags for improved partitioning in aqueous two-phase systems. Qualitatively the peptide-tagged protein partitioning could be predicted from peptide data, i.e. partitioning trends found for peptides were also found for the peptide-tagged proteins. LÄS MER
10. NMR STUDIES ON STRUCTURE AND DYNAMICS OF TWO-DOMAIN PROTEINS
Sammanfattning : The general objective of the present thesis work was to examine 3-dimensional structure as well as the motional properties of two different types of two-domain proteins by solution-state NMR. One of the major tasks of this thesis work was to investigate the domain orientation of these molecules by employing residual dipolar couplings (RDC) which can be used as orientational restraints for increasing the accuracy of protein structures determined by NMR.
