  1. 1. Mutational Analysis and Redesign of Alpha-class Glutathione Transferases for Enhanced Azathioprine Activity

    Författare :Olof Modén; Bengt Mannervik; Mikael Widersten; Kenneth Tew; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; allelic variants; azathioprine; bioactivation; chimeric mutagenesis; directed evolution; DNA shuffling; enzyme engineering; glutathione transferase; GST; lysate screening; molecular docking; multiple alignment; multivariate analysis; polymorphism; principal component analysis; prodrug; prodrug activation; protein engineering; protein redesign; reduced amino acid alphabet; saturation mutagenesis; semi-rational enzyme engineering; site-directed mutagenesis; structure-activity relationship; structure-based redesign; Biokemi; Biochemistry;

    Sammanfattning : Glutathione transferase (GST) A2-2 is the human enzyme most efficient in catalyzing azathioprine activation. Structure-function relationships were sought explaining the higher catalytic efficiency compared to other alpha class GSTs. LÄS MER

  2. 2. Expression of Manganese Lipoxygenase and Site-Directed Mutagenesis of Catalytically Important Amino Acids : Studies on Fatty Acid Dioxygenases

    Författare :Mirela Cristea; Ernst H. Oliw; Chao Su; Olof Rådmark; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical pharmacology; Dioxygenase; Gaeumannomyces graminis; lipoxygenase; Magnaporthe grisea; oxylipin; Pichia pastoris; hydroperoxide isomerase; polyunsaturated fatty acids; Farmaceutisk farmakologi; Pharmaceutical pharmacology; Farmaceutisk farmakologi;

    Sammanfattning : Polyunsaturated fatty acids can be bioactivated by two families of dioxygenases, which either contain non-heme iron (lipoxygenases) or heme (cyclooxygenases, linoleate diol synthases and α-dioxygenases).Lipoxygenases and their products play important roles in the pathophysiology of plants and fungi. 3. 3. Protein Engineering on Azurin. Expression Mutagenesis and Characterisation of Copper Site Mutants

    Författare :B Göran Karlsson; Chalmers tekniska högskola; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; EPR spectroscopy; cassette mutagenesis; optical spectroscopy; amino acid exchange; Escherichia coli; electron transfer; reduction; azurin;

    Sammanfattning : Azurin belongs to a family of small blue copper proteins or cupredoxins which participate in electron transfer processes in plants and bacteria. The type 1 copper site in these proteins is characterised by an intense blue colour, a narrow hyperfine coupling in the EPR signal and, generally, a high reduction potential. LÄS MER

  4. 4. Characterization of galanin receptors using chimeric peptides and site-directed mutagenesis

    Författare :Ulla Eriksson Sollenberg; Ülo Langel; Anna-Lena Hulting; Stockholms universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; galanin; GPCR; mutagenesis; neuropeptide; Neurochemistry; Neurokemi; Neurochemistry and Neurotoxicology; neurokemi och neurotoxikologi;

    Sammanfattning : Galanin, a 29 (30) amino acid neuropeptide, is found throughout both the central and peripheral nervous systems. It signals via three receptors, GalR1-3, all belonging to the rhodopsin-like G-protein coupled receptors. Galanin and its receptors have been implicated in a vast variety of biological processes.
